Radioactive Retinopathy Research Of The Vascular Endothelial Growth Factor In Vitreous And Serum On The Regulation Of Radiation Retinopathy Rat

Objective We use radioactive ray to irradiate SD rats(RR),Observing the content of vascular endothelial growth factor(VEGF)in Serum,vitreous of RR rats.explore the role of VEGF in the RR's development.Methods 45 male Spregue-Dawley(SD)rats model were made by 60Co dioative-ray and randomly divided into normal control group,experiment group. 5%pentobarbital haven been given by Intraperitoneal injection of every rat (0.8ml/100g).After Anesthesia,putting them on irradiation tooling.The radiation field of is 2*2cm..10Gy group:the Absorbed Dose is 0.8/min.It lasts 12.5 minutes.30Gy group:the Absorbed Dose is 0.8/min.It lasts 37.5 minutes. We put the rats to death after collecting the specimens in three months,six months and nine months.After Anesthesia,We use disposable Syringes puncture in 3:00 bell position of Limbal.Collected vitreous about 0.2ML. Preserved in -80℃placement refrigerator.After collecting the vitreous,then cut the ribs in rats exposed the chest.We used the disposable syringes inserted rat left ventricular collecting blood 2M1 placed into a vacuum blood collection tube for centrifugal.After 20 minutes,we collect supernatant about 0.5ml,then immigration and disinfection of the silicide 0.5ml Ep tube sealed Home temperature refrigerator -80℃.The bulbar conjunctiva has been tagged by 5/0′S thread in the eyeball of 12hours place.Dissociating conjunctiva and muscle, shearing the cranial nerve,taking out of the eyeball,putting the left and right eyeball into 10%formalin 24 hours.Removaling protomerite,crystalline,corpotisvitre blow the Operating microscope.Inegativing cutting the sclera of hemispheric with retinal which has been marked.Denuding retinal and made them into trypsin digestion stretched preparation.Use Rat VEGF ELISA kit for detection of VEGF content.Test results are used(x~-)and standard deviation(s) that SPSS 13.0 statistical package using results of univariate analysis of variance and correlation analysis.Results①30Gy rats vitreous concentrations of VEGF:normal control group (77.04±4.36)ng/ml;3 months(84.66±5.97)ng/ml;6 months(316.27±9.62) ng/ml;nine months(663.98±5.93)ng/ml.Three months,six months and nine groups of VEGF concentrations than normal control group Enrichment (p＜0.01).Group 3 months and 6 months group were not significantly different (P＞0.05).Section 9 is the highest concentrations.②10Gy rats serum concentrations of VEGF:the control group(6.78±3.61)ng/ml;3 months(5.73±2.65)ng/ml;six months group(7.09±0.85)ng/ml;9 months group(7.16±2.61)ng ml.Compare with the normal control group,9 months,six months group,three groups of concentration have no statistical difference(P＞0.05).③10Gy rats vitreous concentrations of VEGF:normal control group(70.27±1.65) ng/ml;3 months(72.32±2.16)ng/ml;six months group(75.06±5.55)ng/ml; 9 months group(85.05±4.46)ng ml.Three months,six months and nine groups of VEGF concentrations are higher than the normal concentration of the control group(P＜0.01);Section 9 is the highest concentrations of three months,and the group of six groups showed no difference(P＞0.05).④10Gy serum concentration of VEGF:the control group(5.98±3.64)ng/ml;3 months(4.85±3.16)ng/ml;six months group(6.29±2.40)ng/ml;9 months group(5.11±2.67)ng ml.And the normal control group,9 months,six months or three months in the group were not statistically different(P＞0.05).
