Objective: 1. To study the gene expression of Visfatin in the abdominal adipose of dietary obesity c57BL/6 mice. 2. To investigate the effects of piogalitazone on the Visfatin gene expression.Methods: 1. The weaned male c57BL/6 mice were induced to be obesity with high fat diet for eight weeks. After then, the obesity c57BL/6 mice were sieved out and were randomly divided into high fat obesity group and high fat + pioglitazone group. The weight, Lee's index, visceral fat and abdominal subcutaneous fat weight, fat index, triglycerides, total cholesterol in serum and fasting blood glucose were detected. 2. Expression of Visfatin were detected by fluorescence real-time quantitative PCR.Results: 1. The weight, Lee's index, visceral fat and abdominal subcutaneous fat weight, fat index, total cholesterol in serum and fasting blood glucose in high fat obese mice were higher than the normal controls, except for the triglycerides in serum. And these indices in the high fat + pioglitazone group were higher than the normal controls too, but were lower than the high fat obesity group. 2. Compared with the normal control group, expression of Visfatin in the visceral fat was increased both in the high fat obesity group and the high fat + pioglitazone group, as for the expression of Visfatin in the abdominal subcutaneous fat, the high fat + pioglitazone group was still increased, but not increased in the high fat obesity group. Compared with the high fat obesity group, the expression of Visfatin both in the visceral fat and abdominal subcutaneous fat was increased in the high fat + pioglitazone group. 3 Expression of Visfatin mRNA was positively correlative with body index(r=0.584,p=0.011),visceral fat weight(r=0.676,p=0.002) and body fat index(r=0.704,p=0.001) in the visceral fat. But the Pearson correlation analysis did not show any correlation in Visfatin gene expression and the body weight, or the others in abdominal subcutaneous fat.Conclusion: 1.The dietary obesity c57BL/6 mice model is raised successfully in this paper, which has some major features analogous to human central obesity. 2. Expression of Visfatin mRNA in the visceral fat is very high in dietary obese c57BL/6 mice, which suggests that Visfatin may play an important role in the develpoment of obesity. 3. The insulin sensitizer pioglitazone can increase the expression of Visfatin significantly, both in the visceral and abdominal subcutaneous adipose, which indicates that the pioglitazone may affect the expression of Visfatin, through the way of influence to the body fat distribution.
