Objective To investigate the effect of allogeneic inmmunization on the outcome of pregnancy in patients with unexplained recurent spontaneous abortion, to reseach the percentage of CD4+CD25+ regulatory T cell in patients,peripheral blood before and after allogeneic lymphocyte therapy.Methods 1) The mothod of lymphocyte therapy:infertile women with unexplained recurent spontaneous abortion (n=55) were determined their levels of blocking antibody by using the complement-dependent cytotoxic text, women were subcutaneouly allogeneic immunotherapied with the husbands,T-lymphocyte,and the outcome of pregnancy were followed-up. 2) The proportion measurement of CD4+CD25+ T regulatory cells: The proportion of CD4+CD25+ regulatory T cells in peripheral blood of patients with unexplained recurent spontaneous abortion was measured by double-label flow cytometric analysis.Results 55 Patients with unexplained recurent spontaneous abortion had negative blocking antigen, all the patients recived allogeneic lymphocyte therapy. After immunotherapy 33 patients obtained successful pregnancies, 14 patients abortion again; After allogeneic lymphocyte therapy, The proportion of CD4+CD25+ regulatory T cells in peripheral blood of patients with unexplained recurent spontaneous abortion increased significantly(P﹤0.05), the percentage of CD4+CD25+ regulatory T cells was significantly higher in successful pregnancy women than those in pregnant loss after lymphocyte therapy.Conclusion Allogeneic immunotherapy on patients with unexplained recurent spontaneous abortion would be effective, allogeneic lymphocyte therapy can enhance the proportion of CD4+CD25+ regulatory T cells in peripheral blood, CD4+CD25+ regulatory T cells may play an important role in maintaining materno-fetal tolerance.
