AIM: To investigate the expression of p16, nm23-H1 and p53 in the pathogenesis, infiltration, metastasis, prognosis in GC.Methods: We choose 91 surgical excision of GC in Surgical Oncology of the First Affiliated Hospital of Nan Hua Medical University from 2001-1/2003-1. All the cases were not having all preoperative chemo radiation. They all had completely medical records and we used above mentioned paraffin tissue block. We detect the product of p16, nm23-H1 and p53 gene expression, by immunohistochemistry method of 91 Surgical excision of GC. Test result was divided into two groups, the positive group and the negative group. Statistical analysis was conducted in comparison of data between the data of 91 cases and experiment data.Results: The positive expression of p16, nm23-H1 and p53 in GC were 33.0 %( 30/91), 38.5 %( 35/91), 53.8 %( 49/91). The expression of p16,nm23-H1, p53 were notably corrected with the depth of invasion , lymph nodes metastasis, pathologic type ,lymph nodes, TNM stage in gastric carcinoma(P<0.05).but they not corrected with the age and gender (P >0.05).The expression of p16 ,nm23-H1 were negative corrected with p53 protein (P <0.05). The rank correlation analysis result showed p16 and the nm23-H1 expression is not connected. Also, the expression of p16, nm23-H1, p53 had obvious correction with the Life span of patients (P <0.05). Multivariate Cox proportional hazard model analysis showed that the negative expression with the degree of local infiltration and metastasis of lymph node,TNM different stages, the expression of p16, the expression of nm23-H1 ,the expression of p53 was an independent factor of prognostic significance in GC.Conclusion:1 .There was significant difference among low expression of p16 and nm23-H1 gene in GC tissues , high expression of p53,lymph node status ,infiltration ,Different stages of TNM,and different histology .2. There was inverse correlation between p53 and p16 expression and between p53 and nm23-H1 in GC .There was no relationship between the expression of p16 and nm23-H1 in GC.3.The lower expression of p16, nm23- H1 and increased expression of p53 reflected prognosis of patient with GC, If combined detection of p16 nm23-H1 p53 may be helpful for us to predict the prognosis and useful in treatment of GC .
