Objective To cultivate adult coronary artery endothelial cells in vitro, to studythe effect of valsartan and angiotensinⅡ(AngⅡ) on level of endothelin in culturedhuman coronary artery endothelial cell.Methods Endothelial cells were separated by digesting and scraping fromsudden of death adult coronary artery, they were cultured with Dulbecco's modifiedEagle medium (DMEM). The second generation cells were freezed. The freezingendothelial cells were resurrected and planted into culture board of 6 gabs. The drugswere put in when the third generation cells had reached 60 percent of culture board.They were divided into contrast group, valsartan group (10μmol/l), AngⅡgroup(10μmol/l), AngⅡgroup+valsartan (0.1μmol/l, 1.0μmol/l, 10μmol/l ) group. Wetested endothelin level of cultured human coronary artery endothelial cells after thecells with AngⅡand valsartan had been incubated for 36 hours.Results The adherent rate of cell in primary cultures was 30~40%within 24hours, the cells began to grow at second day; the quantity of cells increased obviouslyat 5th day and the bodies of cell connected tightly; the limitation between cells wasindistinct at 7th day. The cultured cells became confluent monolayer and arrangedclosely at 10th day. The cells in secondary culture that were polygons, had obviousborders and grew actively accorded with the biological characteristic of vascularendothelial cells. Endothelin level were higher in AngⅡgroup than in control group,the difference had meaning of statistics(P<0.000); the difference of endothelin levelbetween valsartan group and control group had no meaning of statistics (P>0.5);endothelin level were higher in AngII group than in AngⅡ+0.1μmol/l valsartan group,the difference had meaning of statistics(P<0.000); endothelin level was higher inAnglI group than in AngⅡ+1μmol/l valsartan group, the difference had meaning of statistics(P<0.000); endothelin level were higher in AngⅡgroup than in AngⅡ+10μtool/l valsartan group, the difference had meaning of statistics(P<0.000); endothelinlevel were higher in AngII+0.1μmol/l valsartan group than in AngⅡ+1.0μmol/lvalsartan group, the difference had meaning of statistics(P<0.000); endothelin levelwere higher in AngⅡ+1μmol/l valsartan group than in AngⅡ+10μmol/l valsartangroup, the difference had meaning of statistics(P<0.000).Conclusion AngⅡincreased endothelin level of human coronary arteryendothelial cells, and the action can be reversed by valsartan. Using alone valsartanhad no effects on endothelin level. The result suggested that valsartan could improvethe function of human coronary artery endothelial cells and prevent atherosclerosis.
