Purpose: The radiation brain injury is one of the serious complications in theradiation.This article choose EPO, the drug for anemia, as a tissue protective drug.Single Cell Gel Electrophoresis (SCGE), Maleic Dialdehyde (MDA), Histopathologylevel investigate the protective action of EPO under the brain radiation injury.Methods: 1) Experimental animal groups, all groups using a random methoddivided into five groups, A: control group, B: irradiation with non-medicine used, C: EPO 150U/kg was given intraperitoneally before irradiation, D: EPO 300U/kg wasgiven intraperitoneally before irradiation, E: EPO 500U/kg was givenintraperitoneally before irradiation. 2) Preparation of the animal model, Anesthesiaagents: Ether. Irradiation method: 6M-X single-brain irradiation, irradiation dose:14.5Gy. Drugs: Name: rHuEPO; Delivery methods: Intraperitoneal administration;Delivery time: half an hour before; half an hour after irradiation; Continuousadministration two weeks after irradiation, three times a week.Observation of the indicators: 1) The effect of EPO on brain after irradiated:Observed the changes of the brain pathomorphology with the Hematoxylin-eosin (HE)staining, observe the effect of EPO to the changes of brain injury.2) The effect of EPOto the free radical after irradiated:Detected the effect of EPO to the contents ofmalondialdehyde (MDA)after the brain tissue homogenate.3) EPO right brainradiation injury circulating blood lymphocyte DNA damage: the single cell gelelectrophoresis from the peripheral blood immediately after irradiation.Results: 1) Compare to the group irradiated with no-medicine used, EPO couldlighten the nerve cell's injury in the after irradiated. 2) EPO could reduce the MDAcontents after irradiated. 3) EPO can reduce circulating blood lymphocyte DNAdamage.Conclusion: The high doses of EPO had protection action in some degree to thebrain radiation injury and had a better effect with pre-injection in our observation.
