Background: Tissues and organs defection or disfunction have been endangered human' s health. While disconnection of bone fracture and bone defection are common diseases in clinical orthopaedics. So that a large amount of medical sources were consumed yearly and many patients were hurt seriously. Nowadays the most principal methods. to treat them are boneautograft, allograft and artificial bone graft. However, all these bones are a kind of 'dead bone' which should be transformed to new live bone by 'creeping substitution' The process is very long and if the bone defection is too large, the new bone could not come into being. And that there are many other limitations of those bones. Bone tissue engineering provides a new method to repair bone defection. Seed cells (osteoblast) is a chief and essential element in bone tissue engineering. Bone marrow stromal cells(BMSC) can be harvested by marrow puncturing easily and can gained comparatively enough cells. The cell culture method is mature gradually and they have clear capability of osteogenesis. So BMSC has been paid more attention all along and been considered the seed ceils that have the most possibility to use in clinic. With adipose-derived stromal cells(ADSC) developing, it has been a research focus because of its stem function. ADSC can be harvested more easily and it is more abundant in human bodies, can be cultured more easily, can proliferate more rapidly. It can be induced to osteoblast definitely so that it can beabetter source of seedcells.Objective: To study the biological characteristic of ADSC and BMSC cultured in vitro and compare their capability of osteogenesis so as to evaluate whether ADSC could be a better seed cells to replace BMSC.Methods: Adipose tissue and bone marrow from a same body were collected during operation. Adipose tissues were digested with collagenase typeâ… to gain ADSC and BMSC were isolated by centrifugation on lymphocyte separation medium. They were all cultured in DMEM/F12 containing volumes of 10% fetal bovine serum. Subsequently, 50mg/L ascorbic acid, 10mmol/L beata-sodium glycerophosphate and 10-8 mol/L dexamethasone were added into culture medium to induce the passaged cells. At the second and third week after inducement, alkaline phosphatase quantity in cells was measured for comparison and used Von Kossa staining to detect the formation of calcium nodus.Results: Adipose tissue and bone marrow were gained from 15 patients and 10 of them brought off a successful experiment. Compared with BMSC, ADSC can be cultured more easily and proliferate faster. Their morphological character were similar. More or less black calcium noduses were seen in extracellular matrix of the induced cells. The alkaline phosphatase quantity between ADSC and BMSC was no significant statistically. The result proves ADSC and BMSC have the alike osteogenetic ability.Conclusion: Bone tissue engineering provids a new method to cure bone defection and fracture disconnection. Adipose tissue is abundant and there are large quantity of stromal cells. ADSC has the alike biological character with BMSC and they can be cultured more easily, proliferate faster, can be induced to osteoblast. The alkaline phosphatase quantity in the cells induced from ADSC and BMSC of one same person was no significant statistically. The result suggests ADSC should be a more ideal seed cells of bone tissue engineering and be more superior than BMSC.
