The Experimental Study Of The Effects Of Verapamil-nitroglycerin Solution On Rabbit Endothelial Cells

Objective:In recent years complete arterial grafts for coronary revascularization is praised highly by more and more surgeries because of its satisfying therapeutic efficacy and long term patency rate.But the injury of endothelial cell and the spasm of grafts are hard to be avoided.Endothelial injury is important element to cause the spasm.The study was expected to observe the effect of verapamil-nitroglycerin solution on rabbit endothelial cells and to provide reference for adopting and improving the storage solutions.Methods:Femoral artery segments were harvested from 16 adult rabbits,which were then stored separately in one of four different storage solutions.The solutions were:(1)verapamil solution(Ver);(2)nitroglycerin solution(Nit);(3)verapamil-nitroglycerin solution(VG);(4)Ringer's solution(Ringer);The samples were examined with immunochemical stain(ⅧR:Ag stain),transmit electron microscopy and specific radioimmunity.To observe the ultrastructure and integrality of endothelial cells and endothelial denudation and the contents of endothelin-1 in vessel wall.Results:(1)Immunochemical stain(ⅧR:Ag stain):vascular endothelial cell appearanced monolayer line.The covering percentage of endothelium monolayer was higher in VG compared with that in Ringer,Ver and Nit(P＜0.05).The percentage in Ringer was lower than others (P＜0.05).There was no statistic difference between Ver and Nit(P＞0.05).(2)The degree of injury was different in different solutions through TEM(P＜0.05).It was significantly lighter in VG compared with Ringer(P＜0.01)and Ver(P＜0.05),but there were no significant differences among the other groups(P＞0.05).(3)The level of endothelin-1 secreting:It was significantly lower in VG than in the rest groups(P＜0.05),while it was higher in Ringer than the other groups (P＜0.05).There was no significant difference among the rest groups.Conclusion:There was different effect in various kinds of storage solutions.Ringer solution might cause more serious damage than the other solutions in terms of the ultrastructure and integrity of the endothelial cells.While verapamil solution and nitroglycerin solution showed some protection in the ultrastructure and integrity of the endothelial cells,but the difference between them was still to be studied.VG solution displayed its superiority on the two directions and it might be a satisfactory solution to be utilized in coronary artery bypass grafting.
