The uterine cervical squamous cell carcinomas is one of the most popular malignant tumors in women.In recent years,the incidence of it in our country is increased obviously and occupies the importment place in malignant tumors.Although the therapy of this cancer is fairly effective,it threatens to women' s health heavily. Now,it's formed that tumors invasion and metastasis is the main reason which results in the patients' death.Participate in the invasion and metastastatic course of the tumor,MMP-7 and MMP-14.They are very important for the degradation of the ECM,and connected with invasion and metastasis.Objective:To study the expression and function relations of the MMP-7 and MMP-14 in uterine cervical squamous cell carcinomas,and compare with normal cervical tissues using TaqMan probes fluorescence real-time PCR.Methods:Materials arc from the patients of Shanxi Province Tumor Hospital,fromMar.2006 to Nov.2006.All the cases are diagnosed by pathologist.Materials we had analysed 30 cases of uterine cervical squamous cell carcinomas,10 cases of normal cervical tissues at contral for the presence of MMP-7,MMP-14 using TaqMan probes fluorescence real-time PCR.The results were analysed for the possible coorelation with disease's outcome.Results:1.Compared with normal tissues,upregulation of MMP-7,MMP-14 mRNA was observed in 30 the cancer tissues beside and cervical cancer tissues t=3.994 P=0.001;t=3.173 P=0.004.2.The expression value of MMP-7,MMP-14 mRNA in FIGO stagingsⅠ,Ⅱ,Ⅲwas increase gradually.Analyzed by one-way ANOVA,there were significantly differences in the expression of MMP-7,MMP-14 mRNA in defferent the FIGO stagings(F=5.107,P=0.009;F=5.806, P=0.008))and grades(F=8.248,P<0.05;F=7.236,P<0.05).Moreover,there were no significantly differences in the expression of MMP-7 mRNA in defferent the FIGO stagingsⅡ,Ⅲand gradesG2,G3(P>0.05).3.The overexpression of MMP-7,MMP-14 was significantly correlated with lymph node metastases.(t=2.268 P=0.033;t=4.454 P=0.000).Conclusion:1.The expression of MMP-7mRNA and MMP-14mRNA in uterine cervical squamous cell carcinomas were significantly higher than in the cancer tissues beside,The expression of those in the cancer tissues beside were significantly higher than in normal cervical tissues.Which suggest MMP-7 and MMP-14 may play an important role in the occurrence,invasion and metastsis of cervical cancer tissues.They may be the valuable clinic marker of cervical cancer with metastassis.2.MMP-7 mRNA and MMP-14 mRNA have significantly relation with the FIGO stagings and grades of cervical cancer.MMP-7 and MMP-14 might be useful for monitoring infiltration and metastasis in cervical cancer.3.The over-expression of MMP-7,MMP-14 was significantly correlated with 1 ymph node metastases.The over-expression of MMP-7 and MMP-14 may play an important role in the invasion and metastasis of cervical cancer,and may be used as a biologic marker in predicting the aggressiveness of cervical cancer.Suppressing MMP-7mRNA and MMP-14mRNA's expression may inhibitor the invasion and metastasis of cancer.4.The cervical cancer tissues inside MMP-7 mRNA's with MMP-14 mRNA's the level of expression,examine to report through the relativity related(r=0.542 t=2.724 P=0.029).
