Objective:To study the expression in rat hepatic fibrosis and leptin in rat blood serum.Methods:40 male Wistar rats weighting about 180~220g were randomly divided into two groups:normal group and bile commen duct ligation model group.The experimental group was induced by bile commen duct ligation.In the 7d,14d,28d,56d of the experiment,rat liver tissues were taken for morphological analysis(by HE stained sections)and the stage of hepatic fibrosis was observed under light microscope.Blood sample were collected to measure serum leptin and leptin was measured using radio immunity technique.The protein expressions of TGF-β1 in liver tissue were determined with immunohistochemistry assay.Results:Morphological analysis showed that the liver function worsened and the liver fibrosis was gradually increased in the bile commen duct ligation induced rats,while the expression of TGF-β1 was increased in liver,compared with those in normal group(P<0.001).There were significant differences in the serum leptin levels at the 7d,14d,28,56day.Conclusion:leptin and TGF-β1 are possibly involved in the formation and development of liver fibrosis.
