ObjectiveTo investigate the expression of BRAF in the serrated neoplasia pathway of colorectal cancer andthe relationship between the expression level and clinic pathological factors.Methods1. 20 hyperplasic polyps, 20 colorectal adenoma (13 lower-moderate dysphasia and 7 severe dysphasia) and 40 colorectal cancers were randomlly collected in The First Clinical Hospital of Shanxi Medical University.2. Immunohistochemistry method was used to detect the expression of BRAF in all groups, the positive signal is that there are brown-yellow grains in the cell liquid.3. Statistical treatment were progressed in SPSS.Results1. In 20 hyperplasic polyps cases, the positive expression rates of BRAF protein was 10.0%. In 20 colorectal adenoma cases, the positive expression rate of BRAF protein was 65% (13 lower-moderate dysphasia, the positive expression rate of BRAF protein was 42.9% and 7 severe dysphasia, the positive expression rate of BRAF protein was 76.9%. In 40 colorectal cancers cases, the positive expression rate of BRAF protein was 80%2. BRAF had a discrepancy in hyperplasic polyps, colorectal adenoma and colorectal cancer (P <0.05). BRAF had a discrepancy between groups of colorectal adenoma.3. The expression of BRAF was correlated to differentiation and lymph node metastasis(P< 0.05),but unrelated to ages of onset and gender(P>0.05).Conclusion1. The expression of BRAF can be found in hyperplasic polyps,colorectal adenoma and colorectal cancers. The BRAF positive expression rate in colorectal cancers is higher than the others.2. In colorectal cancers, there was positive correlation between the positive expression of BRAF with lymph nodes metastasis and pathological types, without correlation with gender and ages.3.The mutation of BRAF was a key chang in early colorectal cancers. The expression of BRAF was closely correlated to the tumorigenesis and progression of colorectal cancer. BRAF appears to be a factor that can be used to instruct the typing of colorectal cancers,also can be used to judge the prognosis of colorectal cancers.
