Chapter1Expression and correlation analysis of BRAF and Caspase-3in gallbladder carcinomaObjective: To investigate the expressions and relationship of BRAF and Caspase-3ingallbladder carcinoma.To analyze the relationship between BRAF and Caspase-3expression andthe clinical pathological features in gallbladder carcinoma.Methods:The expression of BRAF and Caspase-3were detected with immunohistochemistry in69cases of paraffin tissue,including47cases of gallbladder carcinoma,9cases of gallbladderadenoma,13cases of cholecystitis.Results:The percentage of the BRAF positive rate was significatly higher in gallbladdercarcinoma83.0%(39/47) than in gallbladder adenoma22.2%(2/9) and cholecystitis7.7%(1/13). The percentage of the Caspase-3positive rate was significatly lower in gallbladdercarcinoma36.2%(17/47) than in gallbladder adenoma88.9%(8/9) and cholecystitis92.3%(12/13). The expression of BRAF protein in gallbladder carcinoma is not significantly relatedwith patient’s age,sex(,P>0.05). BRAF positive rate were96%,93.3%,96.4%,100%in poorlydifferentiated group, Nevin IV-V group, invasion and metastasis group, higher than the well andmoderateil differentiated group, Nevin I-III group and no invasion and metastasis group68.2%、64.7%、63.2%(P<0.05). The expression of Caspase-3protein in gallbladder carcinoma is notsignificantly related with patient’s age,sex, Nevin and invasion and metastasis (P>0.05),Caspase-3positive rate were63.6%in well and moderateil differentiated group, higher than thepoorly differentiated group12.0%(P<0.05). BRAF and Caspase-3expression in gallbladdercarcinoma is negative correlation,(r=-0.366,P<0.05).Conclusion: BRAF high expression and Caspase-3low expression is associated with gallbladdercarcinoma are closely related. The expression of BRAF in gallbladder carcinoma weresignificantly related with Nevin, degree of differentiation, invasion and metastasis. These resultsshowed that the BRAF plays an important role in gallbladder carcinoma invasion andmetastasis.BRAF and Caspase-3expression in gallbladder carcinoma is negative correlation,These results showed that BRAF and Caspase-3expression may be in coordination with thedevelopment of gallbladder carcinoma. The percentage of theCaspase-3positive rate was significatly lower in cholangiocarcinoma38.9%(14/36) than incholangitis91.7%(11/12). The expression of BRAF protein in cholangiocarcinoma is notsignificantly related with patient’s age,sex(,P>0.05). BRAF positive rate were94.8%,95.0%,94.8%in poorly differentiated group, TNM III-IV group, invasion and metastasis group, higherthan the well and moderateil differentiated group, TNM I-II group, no invasion and metastasisgroup64.7%、60.0%、64.7%(P<0.05). The expression of Caspase-3protein incholangiocarcinoma is not significantly related with patient’s age,sex, TNM, invasion andmetastasis (P>0.05),Caspase-3positive rate were58.9%in well and moderateil differentiatedgroup, higher than the poorly differentiated group21.1%(P<0.05). BRAF and Caspase-3expression in cholangiocarcinoma is negative correlation,(r=-0.472,P<0.05).Conclusion: BRAF high expression and Caspase-3low expression is associated withcholangiocarcinoma are closely related. The expression of BRAF in cholangiocarcinoma weresignificantly related with TNM, degree of differentiation, invasion and metastasis. These resultsshowed that the BRAF plays an important role in cholangiocarcinoma invasion and metastasis.BRAF and Caspase-3expression in cholangiocarcinoma is negative correlation, These resultsshowed that BRAF and Caspase-3expression may be in coordination with the development ofcholangiocarcinoma.
