Objective:The present investigation was designed to explore the protection to retina of a traditional Chinese medicine QingGuangKing and the expression of HIF-1αin retina in rats of acute elevation in intraocular pressure.Methods:90 rats were divided into six groups(15 in each),normal control(A),model control(B),high doze(C),middle doze(E),low doze(F),and Yimaikang(F).All rats in every group were irrigated into stomach by respective medicine for seven days.After that,the rats of group B,C,D,E,F were made into acute ocular hypertension models.The acute ocular hypertension models were established by forcing perfusion of normal saline solution into anterior chamber maintained 60 minutes,and the tension was 120mm H2O.Then each group was irrigated into stomach continuously.And then the rats were euthanatized at 1d,2d,3d,7d,14d after perfusion.Their eyes were enucleated for HE and HIF-1αimmunohistochemistry.Results:1.Morphology results:The level of edema of C-F group was lower than that of model control group(B).2.Immunohistochemistry results:The expression of HIF-1αwas not detected in normal control group.The expression of HIF-1αwas increased significantly in model control group at 1d and then decreased gradually.The expression of HIF-1αin C-F group was significantly different with model control group.Conclusions:1.The increasing expression of HIF-1αin retina in rats of acute elevation in intraocular pressure sustained the theory of hypoxia in damage of optic nerve of glaucoma.2.Qingguangking could decrease the level of edema in retina of acute ocular hypertension model.3.Qingguangking could improve the situation of hypoxia in retina of acute ocular hypertension model.
