Objective:To observe the protective effects of QingGuangAn Granule(QGAG) on retina tissues in rabbit eyes with acute ocular hypertension.Method :32 rabbits were divided into 8 groups at random.One eye was regarded as the experimental group randomly in 4 groups which were treated by perfusing distilled water in stomach,the other eye was regarded as the self-controlling group.One eye was regarded as the treatment group randomly in the other 4 groups which were treated by perfusing solution of QGAG in stomach. The acute ocular hypertension(60mmHg) models were established by perfusing physiological saline in the anterior chamber maintained for 2h in the eyes of the experimental group and the treatment group.The levels of superoxide dismutase(SOD) and malondialdehyde (MDA) in the retina tissue were measured in 1,3,7 and 15 day respectively after the treatment.Meantime,we observed the pathologic changes of retinatissues by transmission electron microscope.Results: The level of SOD in the retina tissue in the exiperimental group was significantly lower than that in the self-controlling group in 1,3 and 7 day respectively after acute ocular hypertension (P<0.01),which was near to the self-controlling group in 15 day (P>0.05). The level of SOD in the retina tissue in the treatment group was significantly higher than that in the exiperimental group in 3 and 7 day after the treatment (P<0.01). The level of SOD in the retina tissue in the treatment group was significantly lower than that in the self-controlling group in 1 day after the treatment (P<0.01), which was near to the self-controlling group in 3,7 and 15 day respectively (P>0.05). The level of MDA in the retina tissue in the exiperimental group was significantly higher than that in the self-controlling group in 1,3,7 and 15 day respectively after acute ocular hypertension (P<0.01).The level of MDA in the retina tissue in the treatment group was significantly lower than that in the exiperimental group in 3,7 and 15 day respectively after the treatment (P<0.01).Electron microscope results showed the discs of the outer segment of visual cells rarefied ,mitochondria in the inner segment swollen,the nerve fiber layer, mitochondria in the retinal ganglion cells swollen.However,the changes described above were markedly abated in the treatment group.Conclusion: QGAG can enhance the activity of SOD and reduced the contents of MDA in the retina tissue with acute ocularhypertention.Thereby,it has the effects of antitoxitation and protecting retina.
