OBJECTIVE To expound the mechanism of YiZhiJianNaoGranule Concintration Fluid (YCF) in treating Alzheimer's disease (AD)systematically by analyzing the differential proteins of entorhinal cortextissues in senescence-accelerated mouse P8 (SAMPS) treated with YCF.METHODS 30 6-month old SAMP8 were randomly dividedinto 3 groups, which are the follows: YCF group (n=10) drenched withYCF, model group (n=10) who were killed without any treatment andcontrol group (n=10) drenched with the same dose of double distilledwater. YCF group and control group were administrated for 8 weeks.Then with the technology of two-dimensional gel electrophoresis (2-DE),we extracted and separated the total proteins of entorhinal cortex tissuesin SAMP8 in the 3 groups respectively. After Coomassie brilliant blue,the images were scanned by ImageScanner and analyzed by Imagemaster2DE analysis software. The diferentialy expressed protein spots wereidentified by peptide mass fingerprint (PMF) based on matrix-assistedlaser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and MSDB and NCBInr database searching.RESULTS1. 2-DE maps of entorhinal cortex tissues in SAMP8 in YCFgroup, model group and control group were established.2. 32 diferentialy expressed protein spots were displayed betweenYCF group and model group, in which 12 proteins were up-regulated, 9were down-regulated in YCF group, and 7 proteins only expressed inYCF group, 4 in model group. There were similar differences betweenYCF group and control group but no manifest differences betweenmodel group and control group.3. 14 diferentialy expressed protein spots such as Aconitatehydratase, NADH dehydrogenase iron-sulfur protein 6, voltage-depend-ent anion channel 1 et al were identified. The functions of these proteinsinvolved in mitochondria function, oxidative stress, neurocyte apoptosisand neuron function according to information supplied in MSDB andNCBInr database, which has a direct bearing on AD. CONCLUSION1. Multiproteins expression of entorhinal cortex tissues in SAMP8can be regulated by YiZhiJianNao Granule, which suggest thatYiZhiJianNao Granule has therapeutic action on AD by multi-targetmeans.2. It is probably the mechanism of YiZhiJianNao Granule intreating AD that YiZhiJianNao Granule plays an important role inmitochondria function improvment, antioxidation, neurocyte apoptosisdecreasing and neuron protection.
