Objective: To study the expressions of PHF-1 and 58K-9 in ischemiareperfusion brain tissue of gerbils and the intervention effect of minocydine.Methods: A cerebral ischemia-reperfusion model in gerbils was established by clamping both common carotids. Fifty gerbils were randomly divided into four groups: normal control group, sham operation group, ischemia-reperfusion(IR) group and minocycline intervention group, in which minocycline(45mg/kg, Bid) were injected from gerbils' abdominal cavity twice a day in the intervention group from the day before experiment.The gerbils were sacrificed in batch and the brain tissue were guickly obtained to make the samples at the 6th hours, lst day, 3rd and 7th day after IR .The expressions of PHF-1 and 58K-9 in brain tissue were detected by using immunohistochemistry technique at the 6th hours, 1st day, 3rd and 7th day after IR. The experimental results were analyzed with Spss13.0.Results: Our experimental results revealed that PHF-1 and 58K-9 were expressed in neurons and gliacytes of the brain tissue in 50 gerbils. There were few expressions of PHF-1 while numerous experessions of 58K-9 in neurons and gliacytes both normal control group of 5 gerbils and sham operation group of 5 gerbils, and there was no significant difference between the two groups (P>0.05). Compared to normal control group, the expressions of PHF-1 in neurons and gliacytes of ischemia-reperfusiong erbils were increased evidently while 58K-9 decreased on the 3rd day and 7th day, especially on the 7th day (p<0.01). The expressions of PHF-1 and 58K-9 on the 3rd day and 7th day in minocycline intervention group were obviously different from the IR group (p<0.01).Conclusions:1. The expressions of PHF-1 in neurons and gliacytes of gerbils were evidently increased following cerebral ischemia-reperfusion which suggest the hyphosphorylation of Tau protein following cerebral ischemiareperfusion and the expressions of 58K-9 were decreased which suggest the damage of the Golgi apparatus.2. The damage of brain tissue following cerebral ischemia-reperfusion can become moderate and the expressions of PHF-1 decrease while 58K-9 increase when intervented with minocycline.
