Objective: To investigate the expression and significance of cathepsin B in retina ischemia reperfusion injury. Methods: Transiently elevating the pressure of the anterior chamber with perfusion of normal saline to induce retina ischemia reperfusion injury, healthy thirty Sprange Dawley(SD) rats were randomly divided into normal control group, ischemia reperfusion group, and sham operated group, rats were killed after reperfusion for 1,6,12,24,72h.Retina histomorphology change and the loss of retina ganglion cells were observed by HE stain, the expression and distribution of cathepsin B was confirmed by immunohistochemistry technique. Results: In normal control group and sham operated group, cathepsin B immunoreactivy is very weak in the retina In ischemia reperfusion group, cathepsin B immunoreacrivity is prominent in retina ganglion cell layer and inner nuclear layer, cathepsin B positive expression was first detected at 1h, and increased at 12h, reached maximal level at 24h(P<0.01), and decreased at 72h.Conclusion: Cathepsin B plays an important role in the pathomechanism of retina ischemia reperfusion injury.
