Epilepsy is a neurological disorder characterized by recurring seizures which are presumed to be the consequence of an abnormal and excessive discharge of a set of neurons in the brain. Epilepsy has multiplex etiological factors, in which drugs are the mainstay of treatment and much attention has been paid to traditional Chinese medicine because the therapeutic effects of antiepileptic traditional Chinese medicine are stabile. In recent years, the roles of NPY have been the hot spot on epilepsy. In this study, I have observed the changes of the ECG and NPY in hippocampus of the tremor rats to study the effects of TCMC on absence-like seizures.Objective1. To observe the changes of ECG and the NPY-positive neurons in hippocampus of tremor rats.2. To compare the difference between TCMC and SV and explore the effects of TCMC further.Methods1. The electrodes were implanted in cortical and hippocampus of rats. The EEG was continuously recorded before and after administration of TCMC.2. The hippocampus was isolated for SP staining to detect NPY positive neurons.Results1. The duration of each absence-like seizures was significantly reduced from 100 % to (44±8) %, (52±18) % and (42±6) % (P<0.01) by low dose, middle dose and high dose at 1 d after stopping administration respectively. The total duration of absence-like seizures in 15 min was significantly reduced from 100 % to (60±18) %, (57±14) % and(57±18) % (P<0.01) respectively.2. Immunochemical results: Compared with normal rats, there are more NPY positive neurons and IOD in hippocampus of the model group. Moreover, NPY positive neurons and IOD in hippocampus of high dose group are more than those of the tremor rat group.Conclusion1. Every dose of TCMC is conspicuously effective for treatment of the duration of each absence-like seizures and the total duration in 15 min on tremor rats.2. Followed by the doses increasing, the antiepileptic roles of TCMC is boosted which is characterized by the powerful drug action and long term duration.3. The augmented express of NPY is highly concerned with absence-like seizures on tremor rats and the antiepileptic roles of TCMC which is perhaps an autospecific protection.
