IntroductionEpilepsy is a common disease of nervous system which character is repeatedly attacked cerebtal function disturbance. Although many anti - epilepsy drugs have been produced, they all have some side effects. Experiment and clinical survey have conformed that tall gastrodia tuber, stiff sildworm, calamendiol, scorpion, Arisacma Consanguineum can rest Wind and spasm, break open phlegm and turbid, smooth liver to latent Yang, calm terror and mind. And they are main components of the traditional Chinese medicine to therapy absence -like seizures and grand mal. Until recently electrostimulating or chemicals are major methods to produce animal models to study anti - epilepsy drugs. However, a lot of data have informed heredity is principal cause of epilepsy. In our study, we use the genetic absence - like seizures tremor rat introduced from Japan as experimental model. After chronic placing electrode in cerebral cortex and hippocampus of tremor rat, we observed the inhibited effects to genetic tremor rat absence - like seizures of tall gastrodia tuber, stiff silkworm, calamendiol, scorpion, Arisacma Consanguineum and their compounds.Experimental and method1. Chronic place electrode in cerebral cortex and hippocampus of tremor ratRats were anesthetized with 1.5% pentobarbital sodium (30mg/kg). Fixthe rat onto the stereotaxic apparatus , cut the skin of the middle between eyesand ears. Scratch out muscles and periosteum , stanch and sanitize. Three poreswere drill at three position( -3mm, 3mm) (3mm, 3mm) ( -2mm, 4mm) to the anterior fontanelle, then placed cortex electrode, reference electrode and hippocampus electrode. Placed the body ground wire subcutaneously at the flank fold and embedded the electrode with dental base acrylic resin powder. Electro-enciphalogram were measured at 7 days after operation.2. Measure EEGRat pro - treatment EEC were measured with multilead physical record at 1 week after operation. Groups A - H were treated orally (10ml/kg) saline, etho-suximide, the traditional Chinese medicine compound boil preparation, tall gast-rodia tuber boil preparation, stiff silkworm boil preparation, calamendiol boil preparation, scorpion boil preparation, Arisacma Consanguineum boil preparation, twice a day for a period of 6 days. EEC was recorded after administration 5-6 days and 1 days after stopping administration. Each was continuously recorded for 15 minutes.Result1. Effects of each group to total duration of tremor rat absence - like seizures in 15 minutesSaline had no obvious effects to total duration of tremor rat absence - like seizures in 15 minutes ( p > 0.05 ) ( see table 1 and figure 2). Whereas compare with that of pre - administration, the traditional Chinese medicine compound obviously inhibit it ( p <0.01) , and after 6 days of administration and 1 days after stopping administration decreased by (60 17)% and (56 18)%. The inhibit effects of ethosuximide in 5 and 6 days decreased by (85 7) % and (74 16) % respectively (p <0.01). Tall gastrodia tuber, stiff silkworm, calamendiol , scorpion, Arisacma Consanguineum administration alone had different degree inhibit effects, after 6 days of administration, decreased by ( 82 9 ) % , (84 11)%, (87 8)%, (88 13)% and(86 5)% respectively (p < 0. 05). Meanwhile, at 1 days after stopping administration, Tall gastrodia tuber, Stiff silkworm, Arisacma Consanguineum decreased by (83 9)% ,(85 10)% and(87 9)%) respectively( p <0.05).2. Effects of each group to duration of tremor rat absence -like seizures Saline had no obvious effects to duration of tremor rat absence - like seizures ( p > 0.05 ) ( see table 2 and figure 3 ). Whereas it was obviously inhibited by the traditional Chinese medicine compound ( p <0.01) , and after 6 days of administration and 1 days after stopping administration decreased by (73 5 ) % and ( 66 4 ) % compared with that of pre - administration.
