Objective: To investigate the correlation between IL-4 gene promoter region +33C/T, IL-13 gene Intron3+1923C/T polymorphisms and asthma, and their impact on total serum IgE levels.Methods: Polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP) techniques were used to determine the IL-4 gene +33 locus and IL-13 gene +1923 locus polymorphisms of 100 subjects in asthma and 80 normal controls respectively. The total serum IgE levels were determined by ELISA in two groups.Results:①There are three genotypes in IL-4 gene, genotype CC, CT, TT. In asthma group the C and T allele frequencies at +33 locus of IL-4 gene were 36% and 64%, 60% and 40% in control group, respectively. There are three genotypes in IL-13 gene, genotype CC, CT, TT. In asthma group, the C and T allele frequencies at +1923 locus of IL-13 gene were 40% and 60%, 62.5% and 37.5% in control group, respectively. Allele frequency in each locus has significant difference in two groups, X~2=16.150, X~2=18.000, P<0.01. The subjects with variant allele have higher risks for asthma than those without variant allele, OR=3.367, OR=2.641, P<0.05.②The genotype distribution in each locus was different in two groups, X~2=20.571, X~2-13.708, P<0.01.③In each locus of the same group, the total serum IgE levels of subjects of variant homozygote, heterozygote were higher than those of subjects of wild homozygote, P<0.01, and the total serum IgE levels have no significant difference in variant homozygote subjects and heterozygote ones, P>0.05.④With the same genotype and locus, the total serum IgE level of asthma group was higher than that of control group, P<0.01.Conclusion:①The +33 locus of IL-4 gene and +1923 locus of IL-13 gene may be one of the important candidate genes for asthma.②Allele +33T in IL-4 gene and allele +1923T in IL-13 gene were associated with asthma.③Variant allele +33T in IL-4 gene and allele +1923T in IL-13 gene were associated with the total serum IgE levels, which induced higher serum IgE levels.
