Objective: To observe the cellularcompatibility of BCBB and PDPB with rabbit bone mesenchymal stem cells, and compare the difference of cellularcompatibility of these two materials. The combine the material with better biocompatibility with BMSCs and transplant it into it bone defects in the radius of rabbit, then compare the therapeutic effect with BCBB combination type I collagen, to provide the experimental evidences for the clinic application.Methods: First, the swine vertebral cancellous bones were boiled, dehydrated,calcined two times then we fabricated BCBB. PDPB were made by the swine vertebral cancellous bones with physical and chemical methods. The cultured rabbit bone mesenchymal stem cells were seeded on BCBB and PDPB in vitro, and the cellularcompatibility of BCBB and PDPB with BMSCs were observed by using SEM and MTT method, then we investigated the optimum time for transplanting. The difference of cellularcompatibility of PDPB and BCBB were compared. We combined the material with better cellularcompatibility with BMSCs and transplanted it into the rabbit bone defects, then compared the therapeutic effect with BCBB compounded type I collagen. After transplanting the BCBB compounded type I collagen into rabbit bone defections, we harvested them in different time and observed them with X-ray and microscope.Results: Four days after composited with BMSCs, cells began to grow on thesurface of PDPB and BCBB, and the rate of cell proliferation reached peak on the sixth and eighth day respectively. Numerous cells were covered on the surface of the material. The cellularcompatibility of BCBB was better than PDPB. After transplanting the BCBB composited BMSCs and BCBB compounded type I collagen into rabbit bone defections, we didn't find the immunological rejection. The function of BCBB composited BMSCs to repairs the bone defection was better than BCBB compounded type I collagen.Conclusion: The optimum time of transplantation for PDPB and BCBBcomposited with BMSCs is cultured on the sixth and eighth day respectively. The cellularcompatibility of BCBB with BMSCs is better than PDPB. The function of BCBB composited BMSCs to repairs the bone defection is better than BCBB compounded type I collagen. BCBB is an ideal scaffold material for bone tissue engineering.
