Objective:To probe the methods of preparing the biphasic ceramicbiologic bone(BCBB)and BCBB compounded BMP and type I collagen, observe the biocompatibility of BCBB with rabbit bone mesenchymal stem cells , investigate the best time to transplant and provide the experimental evidences for the constructing of tissue engineering bone. To discuss the ossification ability of BCBB compounded BMP and type I collagen, we observe the new bone formation of two scaffolds transplanted into the rabbit muscle pouches and bone defects.Methods:The swine vertebral cancellus bones were boiled, dehydrated, calcined two times then we fabricated BCBB and the cultured rabbit bone mesenchymal stem cells were seeded on BCBB in vitro, then investigated the optimum time for transplanting. 4, 8 days later, observed them by SEM. After transplanting the BCBB compounded type I collagen and BMP into rabbit muscle pouches and bone defections, we harvested them in different time and observed them under microscope.Results:BCBB possesses a natural porous structure and good biocompatibility. Its component is similar to the mineral of bone. The pore size of BCBB is (462+123)μm, the pore size of BCBB uneven. BCBB is mainly composed of HA(79. 1%) and TCP(17. 4%), the Ca/P element ratio of BCBB is 1: 1.59+0.11, the average porosity is 59. 5% + 2. 5%. On fourth day, the cells grow in and on the BCBB; on eighth day, the rabbit bone mesenchymal stem cells increase obviously, cells and protuberances overlay the surface of BCBB. Rabbit bone mesenchymal stem cells compounded with BCBB proliferate to a steady stage on the eighth day. After transplanting intothe rabbits muscle bags, BCBB compounded BMP has the ossification ability , there are not obvious immunity responds. The function of BCBB compounded BMP and type I collagen to repairs the bone defection is obvious and the ability of BCBB compounded BMP superior to BCBB compounded type I collagen.Conclusion:BCBB can be prepared by calcined two times and BCBBhas a good biocompatibility with rabbit bone mesenchymal stem cells. The best time to transplant is on the eighth day. The ossification and repairing the bone defection ability of BCBB is obvious, and the ability of BCBB compounded BMP superior to BCBB compounded type I collagen . BCBB is an ideal scaffold material for bone tissue engineering.
