| Objective:To indue the differentiation of rabbit bonemesenchymal stem cells (MSCs) into myogenic cells in vitro and tovalue the transplanted MSCs' survival and differentiation inmyocardiac infarction.Methods: MSCs from rabbit were abstracted, isolated, cultured.Afrer MSCs were induced by 5-azacytidine, MSCs' differentiationwas studied by immunohistochemistry. The model of acutemyocardiac infarction was established by ligating leftantarodecendant arteries (LAD) of 30 rabbits. These rabbits wererandomly divided into three groups: the untransmyocardialrevascularization one (n=10) ;the induced one(n=10) and thetransmyocardial revascularization one(n=10) . The induced MSCswere labeled with 4' 6-dimidino-2-phenylindone(DAPI)andautogenously transplanted into The model of myocardiacinfarction. The transplanted MSCs' survival anddifferentiation were studied 2 weeks after transplantation. Results: The grow of MSCs is adherence to wall. So mast ofHaematooietic stem cells and erythrocyte were excluded throughchanging cultural liquid. MSCs grew rapidly. The MSCs inducedby 5-azsacytidine became more longer and sindle shape andtroponin examined with immunohistochemistry are positive.Through fluorescent microscope, MSCs labeled with DAPI show bluefluorescence. After 2 week of transplantation, the ischemicmyocardium in experiment group can be seen MSCs with bluefluorescence, and array direction of MSCs is similar to that ofcardiac cells. Donor cells derived from MSCs demonstratemyogenic differentiation with the expredssion of troponinT.Conclusion: MSCs are easily gained and grow rapidly. MSCsinduced by 5-azacytidine may develope into myogenic cells andexpress troponinT. DAPI is a better label and the rate of label100%. After 2 weeks of transplantation MSCs transplanted intomyocardium can survive and show myogenic differentiation.
