Objective: To evaluate the function and myocardial interstitial fibrosis of left ventricle of myocardial infracted porcine which cured by autologous bone messenchymal stem cells (MSCs) transplantation by injection of coronary artery, and to discuss the feasibility of this therapy.Methods: The 10 myocardial infarcted porcines which produced by percutaneous coronary embolism were divided into 2 groups: transplantation group, which cured by MSCs; control group, which cured by DMEM; every group has 5 porcines. The bone marrow were drawed from iliac bone of porcine and separated , cultured and purified in vitro, then we could obtain MSCs, and injected it into the coronary artery of myocardial infarcted porcines by cardiac cathete. To observe the change of left ventricular diastole and contraction function 1 and 4 weeks post-transplantation, and to detect the myocardium collagen deposition in infarcted and non-infarcted zone at 4 weeks post-infarcted and post-transplantation. To observe myocardial elementary structure and collagen confluence array by Masson Stain, and to detect the Content of serum MMP-1 and TGF-β1 at fixed time.Result: At 4 weeks post-transplantation, transplantation group had significant improvement in LVSP, +dp/dtmax and -dp/dtmax, meanwhile it's LVEDP decreased (P<0.05 ) ; the deposition of collagen in the infarcted and non-infarcted zone of transplantation group is lower than that of control group(P<0.05) , among which the deposition of collagen in the non-infarcted of transplantation group decreased over pre-transplantion (P<0.05) . Compared with control group, the Content of serum MMP-1 increased significantly at 1 week after MSCs' transplantation, reaching the maximum 2 weeks later(P<0.05) . At the same time , the Content of serum TGF-81 had been decreasing continually until 2 weeks post- transplantation (P<0.05) . Collagen fibers was more well-arranged and its confluence was milder in the infarcted and verge zone of transplantation group compared with control group. Conclusions: (1) The therapy of autologous bone messenchymal stem cells' transplantation was safe and feasible in myocardial infarction, and it can improve evidently the function of left ventricular diastole and contraction.(2) Stem cells' transplantation can reduce the level of TGF-61 in serum, meanwhile accelerate Synthesiss and secretion of serum MMP-1, and it restrains the deposition of collagen in the infarcted zone, decreases it in the non-infarcted zone. So stem cells' transplantation can restrain myocardial fibrosis, reverse myocardial collagen remodeling, prevent collagen fibers confluence, and protect myocardial elementary structure.
