Studies On Chemical Constituents And Bioactive In Valeriana Amurensis Smir .ex. Kom

Valeriana amurensis Smir.ex.Kom belongs to the genus Valerianal L. Valeriana amurensis Smir.ex.Kom is mainly distributed in the Northeast of China and abundant in resource in the Greater Xingan Mountain Region. So far, there is no research work published in V. amurensis although some relative plants in the same genus have been widely used to cure insomnia and other diseases.On the basis of pharmacological action, Valeriana at nervous system, heart blood vessel of brain and resist tumour etc. aspect have got therapeuticaction,up to reseach discover the active constituents of Valeriana at nervous system degenerative diseases AD have got therapeutic action.In order to discover new medicinal plant, we studies Valeriana amurensis Smir.ex.Kom on chemical constituents and pharmacological effect. In this study, the components obtained from the roots and rhizomes of Valeriana amurensis Smir.ex.Kom were elucidated. Besides, the pharmacological effect of the components on AD and their mechanisms of action were examined in vivo and in vitro.The experiment results confirmed that the part of extracted with 50% ethanol from Valeriana amurensis Smir.ex.Kom had the effects of anti-AD. Chemical and pharmaceutical research have been conducted on the part extracted with 50% ethanol from Valeriana amurensis Smir.ex.Kom for the aims of further determining the active compounds and effect mechanism for anti-AD.In this study, isolation procedure was carried out by column chromatography and HPLC; structures of isolated compounds were identified by ¹H-NMR, ¹³C-NMR, ¹H-¹HCOSY, HSQC, HMBC and ESI-MS. Isolation of the 50% ethanol extract of the roots and rhizomes of Valeriana amurensis Smir.ex.Kom afforded 8 lignans compounds, 8 compounds were firstly found in Valeriana amurensis Smir.ex.Kom.Finally, Valeriana amurensis Smir.exKom's the pharmacological action of effective part and its chemical constituents are researched in vitro. in oder to authenticating reliabilty in vivo experiment and filtrating bioactive constituents of the pharmacological action of effective part.Meanwile, we cite a pharmacological action of effective part research way of the Chinese medicine at center nervous system aspect, namely"Chinese medicine cerebrospinal fluid pharmacology", we use cerebrospinal fluid of medicinal ingredient to study cell pharmacology, and combine vivo experiment with vitro experiment, further, proved the effective substances of anti-AD.Moreover, to make experiment condition up to inside condition of inside body efficiency, besides outside body increases medicinal dosage of ten times, and use pharmacological efficiency study of PC 12 cell of ice-dried cerebrospinal fluid.The result indicated, 50%EtOH elucidated part of valeriana amurensis Smir,cerebrospinal fluid of medicinal ingredient and ice-dried cerebrospinal fluid of medicinal ingredient equal vary degree' strengthen cell activity and inhibition cell fade, therefore, proved the effective substances of anti-AD.aboved study offers science basis for studying new modern the Chinese medicine of anti-AD.
