Objective: To investigate the influence of puerarin and berberine, the insulin sensitization traditional Chinese medicine,on the synthesis of male rat testosterone, so as to provide animal experimental evidence for clinical decreasing androgen level of patients with Polycystic ovarian syndrome.Method: 30 healthful male Wistar rats are selected and devided into three groups at random, and every group include 10 rats. After HCG stimulation test, we give rats berberine, puerarin and normal saline to rats in each group respectively by intragastric administration once a day for 4 weeks. And all the rats are put to death on the 29th day and the blood sugar, insulin and testosterone value, finally, didymus tissue is dyed by HE and immune histochemistry reagent to test SF-1 expression taken photo.Result:(1) There is no obvious change in blood sugar and insulin value before and after HCG stimulation test( P>0.05 ), the difference is not statistically significant.(2) The testosterone value in the three groups are all decreased, and the difference of decrease in testosterone value in three groups are not statistically significant (P>0.05) .(3) There is no significant difference of SF-1 positive expression in puerarin and berberine group, the expression of which is obviously lower than that in blank group.Conclusion: Both the insulin sensitization traditional Chinese medicine puerarin and berberine cannot decrease serum testosterone value, but they can influence expression of SF-1.
