Objective: By cell culture technique, the splenic lymphocytes from the control rats and asthmatic rats were isolated and Cultured with different dose NGF/LIF. (leukemia inhibitory factor/nerve growth factor); The expressions of IL-4 and IL-5 mRNA in the splenic lymphocytes were respectively detected by RT-PCR, and the secretions of IL-4 and IL-5 were respectively detected by enzyme-linked immunosorbent assay (ELISA). To disclose the role of NGF and LIF in interaction of Th2 immune imbalance and airway neurogenic inflammation, The paper tries to provide scientific evidence for cell therapy on asthma.Methods: 18 Sprague-Dawley rats were randomly divided into two groups: control group (group A, n=8), asthma group (group B, n=8). The control model was established by inhalation of 0.9% NS, The asthmatic model models were set up by intraperitoneal injection of 100mg ovalbumin and 100mg aluminum hydroxide, and two weeks later received nebulization aspiration of 1% ovalbumin solution. The splenic lymphocytes from the control rats and asthmatic rats were isolated and cultured with exogenously different dose NGF or LIF. The expressions of IL-4 (interleukin-4); and IL-5 (interleukin-5) mRNA in the splenic lymphocytes were respectively detected by reverse transcription-polymerase chain reaction (RT-PCR), and the secretions of IL-4 and IL-5 were respectively detected by ELISA.Results: As compared with the control group, the expression of IL-4 and IL-5 (P<0.05) and the levels of IL-4 and IL-5 (P<0.05) were both significantly enhanced in asthma group, on the other hand, showing exogenously added NGF/LIF respectively upregulated the expression and secretion of the IL-4 and IL-5 in a dose dependent fashion because they were all significantly higher than the values effected by the lower dose NGF/LIF(P<0.05).Conclusion: NGF/LIF may contribute to promote and amplify Th2 cells immune imbalance in asthma by upregulating the expression and secretion of IL-4 and IL-5, one of the Th2 cytoldnes.
