Aims: To investigate the effect of different therapeutic methods and recipes ofTraditional Chinese Medicine (TCM) on expression of nuclear factorκB p65 (NF-κBp65) in hepatic tissue of fatty liver rats and its correlation with the pathological stagein rat fatty liver.Method: The fatty liver rat models were reproduced by feeding with high fat dietcombined with alcohol, and meanwhile soothing liver, invigorating spleen, dispellingdampness, promoting blood circulation, and complex recipes were given to treatdifferent groups respectively for 12 weeks. After the rats were sacrificed, the contentsof serum TC, TG, ALT, AST, etc. were assayed and the histopathological changes ofhepatic tissue were observed by light microscope. The contents of plasma tumornecrosis factor-α(TNF-α) were detected by ELISA method, and the positiveexpressions of NF-κB p65 among different groups by means of immunohistochemicalSP method were detected.Results:1. Compared with uncontrolled group the contents of serum TC, ALT and AST incontrolled group significantly increased (P<0.05). After the intervention of decoction,the contents of serum TC, ALT and AST in each treatment group were lower thanthose in controlled group and there was a significant difference between soothing livergroup and controlled group(P<0.05).2. Light microscope observation: Fatty degeneration from medium to severedegrees in hepatocytes, infiltration of inflammatory cells and much focal proliferationof fibers were found in hepatic tissue of controlled group. Compared with controlledgroup, the histopathological changes in soothing liver group and invigorating spleengroup were significantly ameliorated.3. Positive expression of NF-κB p65 and the steatosis were the lowest in thesoothing liver group and the invigorating spleen group, and enhanced expression ofNF-κB p65 has positive correlation with pathology grades. The contents of plasmaTNF-αwere no statistically significant among the different groups. Conclusion:1. Intemperate diet is the main etiological factors of fatty liver. The functionaldisorders of liver and spleen in TCM theory were the fundamental pathogenesis.2. It is indicated that the therapeutic methods and recipes of invigorating spleenand soothing liver probably have effects of preventing and treating fatty liver throughvarious pathways, such as adjusting lipid metabolism, ameliorating the fattydegeneration of hepatocytes, protecting hepatocytes, ameliorating inflammation, andimproving liver function.3. The high expression of NF-κB p65 in hepatic tissue was maybe the importantpathogenesy of steatohepatitis. The therapeutic methods and recipes of invigoratingspleen and soothing liver can suppress the expression of NF-κB p65 in hepatic tissue. |
