Expression Of PTEN, COX-2, INOS Protein Edometrial Carcinoma And Its Clinical Sinificance | | Posted on:2008-07-31 | Degree:Master | Type:Thesis | | Country:China | Candidate:D M Huang | Full Text:PDF | | GTID:2144360215495504 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective: To study the expression and significance of phosphatase and tensin homologue deleted on chromosome 10(PTEN), cyclooxygenase-2(COX-2)and inducible nitric oxide synthase(iNOS)in endometrial carcinoma and investigate the relation between PTEN,COX-2 and iNOS.Methods: Expression of PTEN, COX-2, iNOS protein were detected by immunohistochemistry method (EnVisionTM method) respectively in 24 cases of atypical hyperplasia of endometrium and 55 cases of endometrial carcinoma and compared with clinical characterristics,such as age, clinical-stage and pathological grade.20 cases of normal endometrium tissues were investigated as control group.Result: 1.The expression situation of PTEN, COX-2, iNOS: The expression of PTEN protein in endometrial carcinoma was significantly lower than in atypical hyperplasia of endometrium and normal endometrium tissues(P<0.05). The expression of COX-2 and iNOS protein endometrial carcinoma was significantly higher than in atypical hyperplasia of endometrium and normal endometrium tissues(P<0.05).2.The relation between PTEN expression and the clinical pathological characters of endometrial carcinoma: The positive rate of PTEN was strongly associated with pathological grade,muscles invasion and lymph node metastasis(P<0.05).There was no relationship between PTEN protein and other clinical pathological characters(P>0.05).3.There was no relationship between COX-2 protein and clinical pathological characters in this experiment(P>0.05).4. The relation between iNOS expression and the clinical pathological characters of endometrial carcinoma: The positive rate of iNOS was strongly associated with pathological grade and muscles invasion (P<0.05).5. The relation between PTEN, COX-2, iNOS: The expression of PTEN was no associated with iNOS(r=-0.157,P=0.252). The expression of PTEN was no association with COX-2(r=-0.051, P=0.711). But the expression of COX-2 was positively associated with iNOS(r=0.363,P=0.007). Conclusion: 1. The expressing level of PTEN in endometrial carcinoma was lower than atypical hyperplasia of endometrium and normal endometrium tissues. The expressing level of COX-2 in endometrial carcinoma was higher than atypical hyperplasia of endometrium and normal endometrium tissues. The expressing level of iNOS in endometrial carcinoma was higher than atypical hyperplasia of endometrium and normal endometrium tissues. All the expressing level of results were highly significant. The results suggest that the expressing changes of PTEN, COX-2, iNOS may activate the oncogene and play an important role in the happen of endometrial carcinoma.2. Low expression of PTEN and high expression of iNOS were related to pathological grade and muscle invasion. The results show that low expression of PTEN and high expression of iNOS may relate with the development of endometrial carcinoma predict poor prognosis.3. The expression of PTEN was no associated with iNOS and COX-2 . The expression of COX-2 was positively associated with iNOS.
