Cervical carcinoma is one of the most common malignant gynecological tumors. Recently, some ingredient extracted from herbal medicine and natural plant is becoming new approaches to prevent and cure tumor. Resveratrol is one of the main ingredients of isoflavones, widely distributed in many dietary plants such as soybeans, memdranous milkvetch root and loded kudzurine root, with many physiological and pharmacological activities. It is regarded commonly that Resveratrol is a potential tumor inhibitor. Although it has studied in various tumors such as leukaemia, breast carcinoma, prostate carcinoma, lung cancer, carcinoma of the large intestine, ovarian malignant tumors, there are few reports regarding its effect and mechanism in cervical carcinoma.Objectives:To study the effects of resveratrol on proliferation, apoptosis and invasion of Hela cells and to study the anti-cancer mechanism of resveratrol.Methods: Hela cells were treated with resveratrol at various concentrations. MTT was used to examin growth and proliferation after treatment with resveratrol; The effects of resveratrol on apoptosis of Hela cells was detected by cytometry and TUNEL;Invasion test were used to observe the effects of resveratrol on invasion ability of Hela cells; In addition, the expression of Survivin, Caspase-3 andβ-catenin protein in Hela cells were analysed by SABC immunohistochemistry.Resultes:1,When the hela cells treated with the reseratrol at the dose of 12.5μmol/L for 24 hours no obviously inhabit effect to proliferation can be seen.2,When the dose of resveratrol up to 25μmol/L and higher, the inhabit effect to proliferation can be seen with a time-dose-depandent.3,When Hela cells brown grain in cell nucleus by TUNEL after treated with resveratrol at the dose of 50μmol/L and higher, in the sight of microscope, obviously apoptosis can be seen. And with a time-dose-dependent.4,Obviously apoptosis can be seen when Hela cells was treated with resveratrol at the dose of 50μmol/L and higher by HE and Gim, in the sight of microscope,. And with a time-dose-dependent.5,More Caspase-3 express and less Survivin express can be seen once the Hela cells investigated with the SABC methoed of immunohistochemistry after the cells has been treated with the resveratrol at the dose of 50μmol/L.6,It can be seen that resveratrol at the dose of 12.5μmol/L actually reduced the invasion of Hela cells by Transwell methoed.7,More express in skin and less express in the inter of the cells ofβ-catenin can be seen once the Hela cells investigated with the SABC methoed of immunohistochemistry after the cells has been treated with the resveratrol at the dose of 50μmol/L.Conclusion1,Resveratrol can obvious inhibit proliferation of HeLa cells once at the effect dose ,with a time-dose depandent.This expreise, when the time going on, the inhibit effect more strong once Res at lower dose, but a smoother increase when the Res at a high dose.2,Resveratrol can induce programmed cell death of HeLa cells by morphological observation and TUNEL. This effect maybe attained by increase the express of Caspase-3; decrease the express of Survivin in Hela cells.3,Res inhibit the invasion of the Hela cells maybe though the methoed that increase the express in skin and decrease the express in the inter of the cells ofβ-catenin.
