Study On The Expression Of Bag-1,Bim,P21～(WAF1/CIP1) And Their Correlations In Endometriosis

Objective: 1 To study changes of the expression of Bag-1,Bim,p21 WAF1/CIP1 in the proliferative and secretory stage in normal endometrium,eutopic endometrium in endometriosis and ectopic endometrium in endometriosis. 2 To study changes of the expression of Bag-1 in normal endometrium,eutopic endometrium in endometriosis and ectopic endometrium in endometriosis. 3 To study changes of the expression of Bim in these three groups. 4 To study changes of the expression of P21 WAF1/CIP1in these three groups. 5 To study the Correlations among these three groups.Methods:1 The study suject:1.1 The study group: 30 patients with ovarian endometriosis who were operated in the gynaecology of the Second hospital of HeBei medical University were selected from Junuary 2006 to July 2006(confirmed by the pathology after operated). 16 patients in the proliferative stage and 14 patients in the secretory stage. We distinguished the menstrual cycle on the basis of last menstrual period and the pathology of the endometrium. Mean age of patients with the study group was 38.1 years old (24 to 45 years old). And this group is divivided two small groups: eutopic endometrium group; ectopic endometrium group.1.2 The control group: 30 patients with ovarian duct coincidence were chose. The endometrium was confirmed to be normal endometrium through pathology. 15 patients in the pro- liferative stage and 15 patients in the secretory stage. Mean age of patients with control group was 29.5 years old (23 to 37 years old). All of the woman in two groups had nomal menstrual cycles; no case history of irregularial vaginal bleeding in recent six months; no case history of eating oral contra- ceptive and fixing IUD in recent six months; case history of treating EMs in recent six months; no case history of hypertension,heart diease, diabete,renopathy,urgent and chronic pelvic inflammatory disease. no other case history.2 Sample collecting: the eutopic endometrium was colleted from the study group on their laparoscopic operatings. At the same time, eutopic endometrium was collected; the normal endometrium was colleted on the clinic service. The specimen was bathed by the PS again and again, fixed by PFA.3 The experiment methods: To determine the expression of Bag-1,Bim and p21 in the normal endometrium,eutopic endometrium in endometriosis and ectopic endometrium in endometriosis byimmuno-histechemical staining. To analyze the results by means of the SPSS11.5. Results:1 the expression of Bag-1,Bim was found predominately in the epithelioglandular kytoplasm on the study group and the control group; while expression of p21 was found predominately in the epithelioglandular kytoplasm and nucleus on the study group and the control group; otherwise, expression of Bim was found in the stroma in some specimen.2 The expression of Bag-1 showed obvious cyclic changes on the control group, the expression of Bag-1 in proliferative stage had significant difference than secretory stage.The expression of Bim was simultan- eously found in proliferative stage and secretory stage on the control group, Its expression in proliferative stage had significant difference than secretory stage.3 To compare the expression of Bag-1 on the control group, eutopic endometrium group and ectopic endometrium group, we found they all had statistically significant: to compare the control group and eutopic endometrium group, X2=4.022 P=0.045; to compare the control group and ectopic endometrium group, X2=15.429 P=0.001; to compare eutopic endometrium group and ectopic endometrium group, X2=4.286 P= 0.038.4 a: Statistical significant was found when we compared the expression of Bim on control group with on eutopic endometrium group, X2= 4.176 P=0.041; b: to compare control group with ectopic endometrium group, X2= 10.549 P= 0.001; c: to compare eutopic endometrium group with ectopic endometrium group X2=1.669 P= 0.196.5 a. Statistical significant was found when we compared the expression of p21 on control group with eutopic endometrium group ,X2= 5.079 P= 0.024; b: to compare control group with ectopic endometrium group, X2= 8.864 P= 0.003; c: to compare eutopic endometrium group with ectopic endometrium group, X2=0.601 P= 0.438.6 On eutopic endometrium group, the expression of Bag-1 was related to that of Bim, P<0.05 r=0.765; the expression of Bag-1 was related to that of p21, P<0.05 r=0.74; the expression of Bim was related to that of p21, P<0.05 r=0.759;On ectopic endometrium group, the expression of Bag-1 was related to that of Bim, P < 0.05 r=0.759; the expression of Bag-1 was related to that of p21, P<0.05 r=0.777; the expression of Bim was related to that of p21, P<0.05 r=0.717.Conclusion:1 Bag-1 and p21 WAF1/CIP1 effected on epithelioglandular cell on the study group and the control group , but did not effect on stromal cell; Bim effected on notonly epithelio- glandular cell but also stromal cell on the study group and the control group.2 In nomal menstrual proliferative stage, the expression of Bag-1 was enhanced; the expression of Bim was decreased, so apoptosis of endometrium was inhibited, reproduction was enhanced; In secretory stage, the expression of Bag-1 was decreased, the expression of Bim was enhanced, endometrium was sheded. Bag-1 and Bim effected on normal endometrium together, controlled its changes of form and function.3 The expression of Bag-1 and Bim showed obvious cyclic changes in normal endometrium, but these changes appeared on the study group.4 To compare the expression of Bag-1 on the control group,eutopic endometrium group and ectopic endometrium group, we found they all had statistically significance, these explained higher expression of Bag-1 was related to the developing of endometriosis, likewise, lower expression of Bim and p21 were related to the developing of endometriosis.5 Bag-1,Bim and p21 WAF1/CIP1 were related to the developing of endometriosis through inter -promotion.
