Objective: Proliferative vitreoretinopathy(PVR) is a serious eye disease,which can lead to blindness,and intricacy and hard curing in clinic.Proliferative membrane were ablated in effect with vitrectomy and retina has been released from membrane, but fundamentality problem-hyperplasia of cells can not be completely settled.Then,it is allimportant to prevent PVR with druggery.Chitosan is favourable in biology and assoiling, and control proliferation of fibroblast.Filing with chitosan in vitreous body had no obvious effect on intraocular structure of eye of rabbit.In order to observe the effect of different concentration chitosian on PVR,change in vitreous body and retina was observed in different time,the content of bFGF and IL-6 was measured with ELISA and RIA.The pathological changes of the retina were observed by optic microscope.And this all will provide experimental reason to prevent PVR with chitosan in clinic.Methods: 40 New Zealand white rabbit were selected,and left eye selected as experiment eyes.The rabbits were divided into five groups in random.Normal group was not injected any medicine in vitreous celom,model group was injected physiol-o g ysalt 0.1ml and 0.1ml blood,experiment 1,2,3 group was injected different concentration chitosan(5mg/ml,10mg/ml, 20mg/ml) 0.1ml and 0.1ml blood.Changes in vitreous body and retina was observed with silt lamp micoroscope,funduscope and BUS.The rabbits were taken out vitreous body,and the wiped off cornea and crystal in anaesthesia on 28th.Obtaining the 10mm×10mm of retina from 2PD distance to optic disc.And fixing the retina in 10% formaldehyde,imbedding it in paraffin to make samples for optic microscope to observe pathological changes of the retina.The content of bFGF and IL-6was measured by ELISA and RIA.SPSS12.0 was used to process all datas.Results: 1 Hyperplasia level of vitreous body and retina in different time by silt lamp micoroscope,funduscope:there was not different significantly between experiment 1,2,3 group and model group on the 1st,3rd,7th day,but the experimental groups were lower than model group.Average level of group was respective2.63,2.25,1.38,0.75on the 14th;the level was 3.00, 2.50,2.07,0.93 on the 21st;and the level was 3.13,2.63, 2.38,1.07 on the 28th;the results of processing by Kruskal-Wallis H, P=0.0047 on the 14th,P=0.0098 on the 21st,P=0.0092 on the 28th,all was significant difference (P<0.01),experiment 3 group was lowest than model group and other two groups (P<0.05),and there was not significant difference between experiment 2 group and experiment 3 group (P>0.05).2 BUS Normal group:the vitreous body was transparent and not abnormity echo;model and experimental groups on the 1 st :finding out of facula of mezzo forte echo of local conglomeration in vitreous body;model and experiment groups on the 3rd:finding out of facula of mezzo forte echo of garrulous,dot and conglomeration shape in vitreous body;on the 7th,model group:finding out light strap of irregular,vary intension mezzo forte echo,and not jointing with eyeball,and sporting obviously in vitreous body,experiment 3 group: finding out facula,of mezzo forte echo of a great deal dot,slice and strip shape and not jointing with eyeball,and sporting obviously in vitreous body; on the 14th ,model group : finding out of facula of mezzo forte echo of wadding,slice and strip shape and not jointing with eyeball,and sporting obviously in vitreous body;experiment 3 group: finding out of facula of mezzo forte echo of dot and strip shape and not jointing with eyeball,and sporting obviously in vitreous body;on the 21st,model group: finding out of facula of assembling flaky mezzo forte echo and light strap of strong echo of strip shape,and not jointing with eyeball and sporting obviously in vitreous body,experiment 3 group:finding out light strap of mezzo forte echo of uneven,different length and irregular shape,and jointing singleness with eyeball but sporting obviously in vitreous body;on the 28th,model group:fining out strong echo of irregular and different intension strip shape,and jointing with eyeball and lcking sporting in vitreous body,experiment 3 group:finding out facula of mezzo forte echo of irregular shape,and not jointing with eyeball and sporting obviously. 3 The content of bFGF,IL-6 in vitreous body3.1 Compared with normal groupThe content of bFGF in the model group was (282.20±32.88) pg/ml,that was increased significantly compared with normal group(86.29±11.05)pg/ml(P<0.01);the conten in the experiment groups was respectively(231.01±50.79)pg/ml,(145.79±33.77) pg/ml,(123.45±25.14)pg/ml,by statistic,experiment 1 and expe- riment 2 were compred with normal P<0.01,experiment 3 was compred with normal P<0.05;the content of IL-6 in the model group was(295.97±106.40)pg/ml,that was increased significa- ntly compared with normal group(37.97±4.66)pg/ml,P<0.01;the conten in the experiment groups was respectively(230.08±51. 28)pg/ml,(169.31±42.07)pg/ml,(116.83±17.04)pg/ml,by statist- ic,all that was increased significantly compared with normal group,by statistic,P<0.01.3.2 Compared with control groupAll the conten of bFGF in the experiment groups was decreased significantly compared with control group,by statistic,all P<0.01; all the conten of IL-6 in the experiment groups was decreased significantly compared with control group(experiment 1 group,P<0.05,experiment 2group and expe- riment 3 group,P<0.01)3.3 Compared between groupsThe content of bFGF in the experiment 2 and 3 was different significantly compared with experiment 1(P<0.01),but there was not significant difference between experiment 2 group and experiment 3 group(P>0.05);the content of IL-6 in the experiment 2 group and 3 group was different significantly compared with experiment 1(experiment 2 group P<0.05, experiment 3 group,P<0.01);but there was not significant differ- ence between experiment 2 group and experiment 3 group (P>0.05).4 Histopathology change Normal group:the structure of retina was in focus,not dropsy and the number of cells did not change,and viteous body was transparent,did not effuse,bleed and form fibre film;model group:nerve fiber layer occured to dropsy,the layer of ganglion cell was gentle dropsy,the number of cells decreased and the number of glia cells increased,it occurred to a little effusion and blood and form fibre film,and rotundity and shuttle cells in the vitreous body;the structer of retina in experiment 1 group was in turbulence,it occurred to local dropsy in the nerve fiber,and effuse and bleed in the vitreous body,and form fibre film; the structer of retina in experiment 2 group was in turbulence,it occurred to local dropsy in the nerve fiber,and fibrin effusion in the vitreous body,and form fibre film;the structure of retina in the expetiment 3 group was gently in turbulence,and cells were in order, it occurred to local dropsy in the nerve fiber,and there has a little effusion attached to inner membrane.Conclusion:1 All the content of bFGF and IL-6 in the model group was increased significantly compared with normal group.It suggests that bFGF and IL-6 were participated in the process of proliferative vitreoretinopathy in happening and developing .2 All the content of bFGF and IL-6 in the experiment groups was decreased significantly compared with model group.It suggests that Chitosan can decrease the content of bFGF and IL-6 in the vitreous body,and then prevented the proliferative vitreoretinopathy.3 It was inverse ratio relation between the concentration of chistosan and the content of bFGF and IL-6,and then in a certainty,hoisting the concentration of chitosan can reinforce the effect of preventing PVR.4 Chitosan has favorable foreground as a medicine injected into vitreous body in preventing PVR in clinic.
