Objective:1.To analyse the relationships of serotonin transporter(5-HTT)gene polymorphisms and major depression(MD).2.To analyse the relationship between social psychic factors and MD.3.To investigate the influence of interaction effect between 5-HTT gene polymorphism and social psychic factors on MD.Methods:A case-control sample,including 239 patients and 239 controls,was recruited in our study with strict selected standards.EPQ,LES,SSS and TCSQ were used to evaluate the social psychic factors,such as personality,life events,social supports and coping styles.PCR was used to detect the polymorphism of 5-HTTLPR and Stin2.The genotype and allele frequency distribution were compared with UNPHASE software.Multivariate logistic regression was used to select social psychic factors and complete interaction.Results:1.The result of the relationship between 5-HTT gene polymorphism and MD①There were no statistical differences of genotype and allele frequency between patients and controls,and the OR values were not get statistical significance yet.②There were no significant differences of the distribution of STin2 genotypes and alleles between patients and controls,and the OR values were not get statistical significance yet.③Haplotype analysis between 5-HTTLPR,STin2 showed that they belonged to different block and should be managed separately.2.Results of association between social psychic factors and MD①All social psychic factors were risk factors of MD,OR values were 2.416,5.783,10.808, 2.591,3.517,9.492,2.468 seperately②Results of interaction effect between social psychic factors showed:Every social psychic factor had interaction effect with each other.Negetive coping style had rivalry effect with other social psychic factors,indicated that co-effect of them could decrease MD onset.While other factors had synergistic effect of MD.3.Results of relevance between interaction effect of 5-HTT gene polymorphism and social psychic factors and MD①The relationship of MD with interaction of 5-HTTLPR and Social psychic factors There were significant interaction between 5-HTTLPR SS genotype and psychotics personality,less positive coping style and low social supports,γvalue were 1.387,1.329,1.150, showed they could increase MD morbility.②The relationship of MD with interaction of STin2 and social psychic factorsThere were significant interaction between STin2.12/12 and psychotics personality,low social supports,γvalue were 1.337and 1.277 showed they could increase MD morbility.There were also significant interaction between STin2.12/12and introverts,γvalue was 0.847 showed they could decrease MD morbility.Conclusions:1.The 5-HTT polymorphism is not associated with the morbility of MD in Chinese Han.2.All social psychic factors can raise risk of its morbility.Negetive coping style had rivalry effect with other social psychic factors,indicated that co-effect of them could decrease MD onset.While other factors had synergistic effect of MD.3.In Chinese Han population,There were interactions between 5-HTT gene polymorphism and personality,life events,coping style and social supports,5-HTT gene have cooperative or antagonistic influence of social psychic factors and MD morbility.
