Study On Relationships Among The Infection Of Human Papillomavirus 16/18,Human Herpesvirus 6 And P53 In The Laryngeal Squamous Cell Carcinoma

Objectives:Highly oncogenic human papillomaviruse 16/18(HPV16/18)are important, but not sufficient for the development of laryngeal squamous carcinoma.HHV6 has been detected in some neoplasms.To illuminate the pathogenesis,this study examined the possible roles of HPV16/18,HHV6 and p53 at the level of protein and gene in the laryngeal squamous carcinoma,and provid a trend for curing and preventing the laryngeal cancer in theory,using gene therapy in the molecular biology.Materials and Methods:1.Paraffin imbedding tissues:40 laryngeal squamous carcinoma tissuses,30 adjacent normal tissues and 30 polyp of vocal cord tissues were obtained from operated patients,in which HPV16/18,HHV6 and p53 were detected by immunohistochemistry SP.2.Fresh or frozen tissues:30 laryngeal squamous carcinoma tissuses,25 adjacent normal tissues and 25 polyp of vocal cord tissues were obtained,in which HPV16,18mRNA were detected by the reverse transcription polymerase chain reaction(RT-PCR);HHV6 was detected by the the nested RT-PCR.3.Finally statistical calculations were done to analyse the associations of HPV16/18, HHV6,p53 and the clinical parameters and clinicopathological features.Results:1.The average protein of p53 have difference between the LSCC tissues and adjacent normal tissues/the polyp of vocal cord tissues(P＜0.05).The average protein of HPV16/18 have obvious difference between the tissues of LSCC and adjacent normal tissues/the polyp of vocal cord tissues(P＜0.001);The average level of protein of HPV16,18 mRNA have obvious difference between the LSCC tissues and adjacent normal tissues(P＜0.001),have obvious difference between the LSCC tissues and the polyp of vocal cord tissues(P＜0.05).Obvious correlation existed between the average level of protein and mRNA of HHV6 between the LSCC tissues and adjacent normal tissues/the polyp of vocal cord issues(P＜0.001).2.There have difference in the grade of patho-differentiation of p53 and HPV16/18(P＜0.05),the lower of the degree of patho-differentiation,the much of the contents.There have difference in the metastasis of lymph node of p53(P＜0.05),the metastasis have much contents. There was no significant association between HHV6 and age,sex,clinical classification,T stage, N stage,grade of patho-differentiation and the type of the LSCC(P＞0.05)。 3.Obvious concordances existed between HHV6 and HPV16/18 in tumor tissues(P＜0.005), and there have strong concordances to each other.And there have correlations between the levels of HHV6,HPV16/18 and p53(P＜0.001)Conclusions:1.The infection of HPV16/18 may mediate and promote the abnormal control of p53,which play important roles in the development of the laryngeal squamous carcinoma.2.HHV6 takes part in the development of malignancy and the cancerization of the cells,it may have associations of p53.3.The infection of HHV6 have close relation to the development of the laryngeal squamous carcinoma,it may be a kind of causative agent.In addition,as a promoter it may participate in and/(or)up-regulate HPV 16/18 urging the laryngeal squamous carcinoma to develop.
