OBJECTIVE: To provide experimental data for clinical therapy bydetecting the antibiotics resistance of staphylococcus aureus after livertransplantation. To understand the infection of MRSA by detecting mecAgene. To provide scientic evidence for preventing, decreacing andcontrolling hospital infection by analyzing the genetic homology ofMRSA.METHODS: MIC was performed using agar dilution method.MecA gene was detected by polymerase chain reaction (PCR). Therandom amplified polymorph DNA (RAPD) was used to identify thegenotypes of strains.RESULTS: Among 34 strains of staphylococcus aureus, thepercentage of resistance accounted for over 60%to six drugs: Oxacillin,Cefoxitin, Meropenem, Pazuflosxacin, Amikacin and Minocycline; 26strains were resistant to five or more drugs. None of the isolates showedresistance to vancomycin and teicoplanin. The isolated rate of methicillin—resistant Staphylococcus aureus (MRSA) was 94.1%(32/34). Allstrains of MRSA were positive to mecA gene. The numbers of amplifiedDNA fragments by RAPD were between three and nine. They wereclassified into 14 genotypes. The main genotype of MRSA isⅢin ourhospital.CONCLUSIONS: The staphylococcus auerus especially MRSAafter liver transplantation have high and multi drug-resistance. We shouldselect reasonable antibiotics in clinical therapy. Detection of mecA geneby PCR is an exact and rapid approach to identify MRSA. RAPD showspowerful discrimination, high typeability, good reproducibility, simplicity,rapidness and economic. It can be widely used to analyze thehomogeneity of MRSA. There is no direct relationship betweenphenotypes divided by antibiotic resistance and genotypes by RAPD.
