CE has several advantages: rapid and high resolving power, automation, low consumption of sample, solvent and chiral selector, as well as its high flexibility in choosing and changing types of selectors and operate modes. It is increasingly developed to become an important technique used in separation. Recently, it has been applied to a wide range of reseach fields such as biology, chemistry, pharmaceutical analysis, environmental protection and foods science et al. In this paper, fingerprints of Momordica grosvenori Swingle, Zanthoxylum nitidum (Roxb.) DC., Phellodendron chinense Schneid. and Sophora tonkinensis Gapnep. and the internations between three alkaloid compounds to HSA were studied by CE.This paper contains two primary parts: First, the capillary electrophoresis fingerprint (CEFP) of Momordica grosvenori Swingle, Zanthoxylum nitidum (Roxb.) DC., Phellodendron chinense Schneid.and Sophora tonkinensis Gapnep. were established to evaluate the quality of four Chinese traditional medicines by CE. Sencod, the binding constants (K_b) of three alkaloid compounds with human serum albumin (HSA) were determined by capillary zone electrophoresis (CZE). The first part includes charpter 2,3,4 and charpter 5, and the second part includes chapter 6. And the main contents of it are as follows:1. The capillary electrophoresis fingerprints (CEFP) of Momordica grosvenori Swingle was established to evaluate the quality of Momordica grosvenori Swingle by CE,with the detection wavelength of 203 nm, applied voltage of 20 kV and detection temperature of 20℃. The borate buffer (adjusted to pH 10.5 with sodium hydroxide) containing 15% acetonitrile and 13 mmol/L SDS be used as running buffer. 26 possessing peaks were selected as the fingerprints of Momordica grosvenori Swingle, and the quercetin was selected as the reference peak. The method presented has good precision, reproducibility and stability, which can be used for the quality control of Momordica grosvenori Swingle.2. The NACEFP of Zanthoxylum nitidum (Roxb.) DC. was established to evaluate the quality of Zanthoxylum nitidum (Roxb.) DC. by CE, the background electrolyte (BGE) was a 100mmol/ L sodium acetate trihydrate (adjusted to pH 5.0 with H3PO4 solution) .The detection wavelength was 228 nm, the voltage of 20 kV and temperature of 18℃was applied.14 possessing peaks were selected as the fingerprints of Zanthoxylum nitidum (Roxb.) DC., and chelerythrine chloride was selected as the reference peak. The CEFP has good precision, reproducibility and stability, which can be used for the quality control of Zanthoxylum nitidum (Roxb.) DC..3. The NACEFP of Phellodendron chinense Schneid. was established to evaluate the quality of Phellodendron chinense Schneid. by CE, with the detection wavelength of 210 nm, applied voltage of 25 kV and detection temperature of 20℃. The 40 mmol/L sodium acetate and 40 mmol/L ammonium acetate methanol solution (adjusted to pH 5.8 with 36% acetic acid) were used as BGE. 13 possessing peaks were selected as the fingerprints of Phellodendron chinense Schneid., and the nitidine chloride was selected as the reference peak. There were good similarities between the standard NACEFP and each NACEFP of samples collected from ten different places. The coefficients were from 0.718 to 0.982. The method presented has good precision, reproducibility and stability with the RSDs of the relative migration time less than 1.26% and the RSDs of the relative peak area less than 3.82%, which can be used for the quality control of Phellodendron chinense Schneid..4. The NACEFP of Sophora tonkinensis Gapnep was established to evaluate the quality of Sophora tonkinensis Gapnep by CE, with the detection wavelength of 210 nm, applied voltage of 25 kV and detection temperature of 20℃. The 100 mmol/L sodium acetate of 80% methanol solution (adjusted to pH 4.8 with acetate acid) were used as BGE. 11 possessing peaks were selected as the fingerprints of Sophora tonkinensis Gapnep, and the berberine was selected as the reference peak. The coefficients were from 0.870 to 0.984.The NACEFP has good precision, reproducibility and stability with the RSDs of the relative migration time less than0.80%, and the RSDs of the relative peak area about 4.12%, which can be used for the quality control of Sophora tonkinensis Gapnep.5. The binding constants (Kb) of three alkaloid compounds with HSA were determined by capillary zone electrophoresis (CZE) with the detection wavelength of 210 nm, applied voltage of 20 kV and temperature of 36.5℃. The phosphate buffer ( pH 7.4, 10mmol/ L ) was used as running buffer. The binding constants of three alkaloid compounds with HSA could be measured by the change of migration time. The experimental values of K_b were 8.785×10~4,7.081×10~4,8.085×10~4 L/mol. The method presented has easy, reproducibility, meanwhile the method has feasibility by comparing to fluorescence. |
