Objective: To investigate the expression and implications of COX-2 in bone marrow cells of acute leukemia patients, and reveal the potential pathogenetic that COX-2 in bone marrow cells of acute leukemia patients.Methods: Western blot was used to measure the expressions of Cox-2 and Bcl-2 in bone marrow cells of 67 cases of acute leukemia before and after treatment with chemotherapeutics.Results: The positive rate of COX-2 in bone marrow cells of acute leukemia was 53.73%(36/67).The COX-2 level in 28 primary patients with primary leukemia was 0.5627+0.0756, significantly higher than that in control group(p<0.05); and was reduced to 0.1851±0.0511 in patients with complete remission, no significant difference composed to control group. The COX-2 concentration of the 8 replase patients was 0.3527±0.1634 and 0.5480±0.1082 in complete remission phase and after replase respectively, the difference was significant(p<0.05). The COX-2 level was related to the Bcl-2 level(r=0.937, P<0.05) and proliferative degree of bone marrow, but not related to the percentages of leukemia cells in the bone marrow, hemoglobin concentration and platelet count. The white blood count(WBC) in group of COX-2 positive and group of COX-2 negative were 69.30±56.83(*10~9/L) and 34.10±12.77(*10~9/L) respectively, the difference was significant(t=3.6123, P<0.05). The LDH concentration n group of COX-2 positive and group of COX-2 negative were 358.96±99.03(IU/L) and 295.25±113.44(IU/L) respectively, the difference was significant(t=2.1684, P<0.05). Conclusion: The level of COX-2 in primary patients with acute leukemia was much higher than that in control group; it was reduced to the normal level after complete remission. It rised again when replased. The level of COX-2 was well corresponded with Bcl-2,WBC,LDH and proliferative degree in bone marrow. It was significant to detect COX-2 concentration in the diagnosis, evaluating disease condition, predicting replase, assessing prognosis, and directing therapy of acute leukemia.
