Objective: This topic mainly takes the traditional chinese medicine theory "in the search for liquid begin" as a breakthrough and syndrome of spleen qi deficiency as a starting point, from the aspect of salivary metabonomics to study the scientific connotation of syndrome of spleen qi deficiency, which is expected to permit the diagnosis of biomarkers with new discoveries.Method: With the method of combining disease identification and differentiation of symptom-complex, people suffered from chronic gastritis or peptic ulcer disease and syndrome of spleen qi deficiency accepted as the experimental group, normal people as a normal control group, patients of chronic gastritis or peptic ulcer disease and syndromes of damp and heat stagnating in spleen as experimental control group. Stimulated whole saliva specimens were collected for a 5-min period using a cube of gum base as a stimulant, according to standardized collection proceures, then spit outside the saliva secreted during the period into aseptic sputum Cup. saliva samples were collected from 9:30~10:30 AM in a quiet room, Saliva was clarified by centrifugation at 14,000 g for 20 min at 4℃and then tested immediately or stored at -70℃until use. PH was measured using saliva pH meter, using automatic biochemical analyzer to test saliva biochemical indicators, Using nuclear magnetic resonance (NMR) method in saliva 1H NMR spectra, then carries on centralization and the scale adjustment for its integral valve, principal components analysis (PCA) by application of Simca-P 10.0 software to study the metabolic characteristics of saliva.Result: (1) Comparing with the normal control group, Group of pleen qi Deficiency Syndrome salivary pH and biochemical indicators of potassium (K) and chlorine (C1),alanine aminotransferase (ALT),aspartate aminotransferase (AST),total protein (TP),albumin (ALB),globulin (GLB),albumin and globulin ratio (ALB/GLB),creatinine (Crea),uric acid (UA),lactate dehydrogenase (LDH),creatine kinase (CK),glucose (GLU) and triglyceride (TG) levels have no statistical significance (P>0.05). Syndrome of Spleen qi Deficiency Group saliva calcium (Ca),magnesium (Mg),phosphorus (P) and blood urea nitrogen (BUN) are significantly higher than that of the control group (P<0.05)Salivary sodium (Na) is significantly lower than the control group (P<0.05).syndromes of Damp and Heat Stagnating in spleen group saliva magnesium (Mg),phosphorus (P),aspartateaminotransferase (AST) and triglyceride (TG) levels are significantly higher than the control group (P<0.05), TG content ofwhich is significantly higher than that of spleen qi deficiency syndrome group (P<0.05).(2) The integral value of main ingredients of three groups saliva typical IH NMR spectra concentrated in three regions of the oval scatter (95% Confidence region), group of spleen qi deficiency syndrome, syndromes of Dampness and Heat in Spleen group and the normal control group have no intersect and overlap; syndromes of Dampness and Heat in Spleen group concentrated in the far left, spleen qi deficiency syndrome group concentrated in the upper right hand corner, and the normal control group concentrated in the lower right corner. Further analysis for the two principal components :PCI and PC2 ,which may affect the IH NMR spectra significantly showed ,in comparision with the normal control group, the salivary lactic acid (1.34 ppm),N-acetyl glycoproteins (2.1 ppm),ethanol (1.18 ppm) content of spleen qi deficiency syndrome group are relatively high. syndromes of Dampness and Heat in Spleen group mainly acetate (1.9 ppm and 1.94 ppm), propionate (1.06 ppm), alanine (3.78 ppm) content are relatively high. This suggested the salivary metabolic network are obvious different among syndrome of spleen qi deficiency,syndromes of Dampness and Heat in Spleen and normal group.Conclusion: saliva metabolism spectrum of syndrome of spleen qi deficiency significantly deviated from the normal crowd, indicating that the Chinese medicine theory "in the search for liquid begin" is scientific.
