In our systematical study on the chemical constituents of Penicillium oxalicum, 20 compounds were obtained from the acetone extract of the mycelia through various chromatographic techniques. MTT assay was used to evaluate the bioactivities in vitro.On the basis of physico-chemical property and spectroscopic analysis, Their structures were elucidated as cyclo-(Phe-Ile)(1), cyclo-(D-Phe-D-Val)(2), cyclo-(Val-Val)(3), cyclo-(Ile-Leu)(4), cyclo-(Pro-Val)(5), 1-hydroxy-1,3,3-trimethylbutyrolactam (6)~*, cyclo-(Pro-Tyr)(7), 3-isopropyl-6-ethylpiperazine-diketopiperazine (8), cyclo-(Leu-Ala)(9), cyclo-(Pro-Gly)(10), Daidzein(11), Genistein(12), Hexadecanoic acid, 2,3-dihydroxypropyl ester(13), Octadeca-9, 12- dienoic acid 2,3-dihydroxy-propyl ester(14), D-mannitol(15), p-hydroxy benzene acetic-acid (16), Adenosine (17), Uracil (18), 2'-deoxyuridine(19), Thymidine(20) respectively. Compound 6 is a new natural product. Compounds 1-4, 7-10 were isolated for the first time from the Penicillium genus. The absolute configuration of compound 2 was determined based on chemical correlation.The cytotoxic activities of compounds were evaluated by MTT method. The results showed that compounds 2, 4, 8 displayed cytotoxic activity in vitro on different levels. On the basis of MTT assay, the structure-activity relationship was discussed. The isopropyl in the structure seemed to play an important role in the activity.
