| Objective: 1,To measure the half lethal dose(LD50 ) and to study thehistopathology for the leaves of the que-zui-cha in mice. 2,Extraction,isolation and identification are studied on the chemical constituentsfrom que-zui-cha. 3,Seven samples are studing to evaluate the effecton blood glucose in mice with diabetes.Methods: 1,Totally 50 mice in half genders were randomly divided into5 groups. Each group had 10 mice . Feeding the mice with different volumeof the que-zui-cha, the toxic symptom was observed. Then calculate theLD50 of the que-zui-cha. 2,The que-zui-cha was extracted with ethanolto get condensate. The condensate was diluted by water then extracted withpetroleum,acetic ether and n-butanol respectively. Acetic ether phasewas chromatographied with silica gel column. Using MS, 1HNMR, 13 CNMRdetermined the structure of the compound that we got from the que-zui-cha.3,ICR male mice (24-26g) were divided into two groups randomly. Thecontrol group (group A)was fed on normal diet, while the other mice wereinjected in 60mg. kg-1 alloxan from tails and fed on normal diet. The mice with blood glucose above 11.1 mmol/L were chosen as diabetic mice. Thediabetic mice were randomly divided into 8 groups according to their valueof blood glucose and body weight. Group B(the negative group),GroupC(hong-xue-cha),Group D(di-sheng),Group E(mi-die-xiang),GroupF (deng-zhan-hua),Group G (he-shou-wu),Group H (que-zui-cha),GroupI (yunnan-shan-zha),Group A and ? were i. g. N. S. The experiment lasted 2weeks. Every week the blood glucose was measured. 4,We go on nextexperiment with more mice and extension dose in same way to evaluatemi-die-xiang,he-shou-wu and que-zui-cha, Group number is group 1 (controlgroup ),group 2 (negative group )…Results 1,The mice had toxic symptom after fed the condensate. TheLD50 of the que-zui-cha is 39.9g/kg. 2,We got two compounds fromacetoacetate phase. Number 1 is hydroquinone and 2 is p-hydroxyphenyl6-0-tans-caffeoyl-β-D-glucoside. 3,We express all the index with(?)±S and statistic all the data with spss 12.0.Compared group B withother group respectively, the difference was significant of que-zui-chaand he-shou-wu. It showed that they had the effect of decreasing bloodglucose. 4,I. g. for 1 week, Compared group 2 with other grouprespectively, the difference was significant except low dose ofhe-shou-wu (p<0.01 or p<0.05) , which shows that they had the effect ofdecreasing blood glucose. After 2 weeks later irrigated, the differencewas significant about group mi-die-χiang and he-shou-wu (p<0.01 or p<0.05) .Conclusion 1,The que-zui-cha has slight toxicity to heart,enteron,brain,liver and so on. 2,Compound 1 and 2 were isolated firstlyfrom que-zui-cha . There is high content about the number 2 compound. 3,que-zui-chaN he-shou-wu and mi-die-xiang can decrease the blood glucose . |
