Objective: To investigate the expression of tissue Transglutaminase in renal tissue of patients with the chronic renal disease including the Lupus Nephritis (LN),proliferative sclerosing glomerulonephritis (PSGN),IgA Nephropathy,Nephritis of anaphylactoid puerperal and their relationship with clinical manifestation. Methods: The expression of tTG in 15 renal biopsy specimens of patients with LN,7 specimens of patients with PSGN,9 specimens of patients with IgAN,6 specimens of patients with Nephritis of anaphylactoid puerperal were detected by immunohistochemical stains. We also examinated the relationship between the expression of tTG in renal tissue and possible associated clinical datus. Results: 1) there is very few expression of tTG in nomal glomeruli and renal tubular-interstitial. Compared with the normal control group , it is significantly higher that the expression of tTG in renal biopsy specimens with LN group,PSGN group,IgA group,and Nephritis of anaphylactoid puerperal group. (P<0.01) 2) Compared with the LNⅡgroup, it is signigicantly higher that the expression of tTG in LNⅣgroup. (P<0.01) 3) There are no significantly differences of expression of tTG in renal biopsy specimens with LN group,PSGN group,IgA group,and Nephritis of anaphylactoid puerperal group. (P>0.05) . 4) In LN group,IgA group and Nephritis of anaphylactoid puerperal group, the expression of tTG in renal tissue are collection with the TMP of the patients. In the LN group, the expression of tTG in renal tissue is collection with C3,C4 and the pathological change of the patients. ConcIusion:This study has demonstrated that the overexpression of tTG in the renal tissue of human chronic renal disease.The more severity of renal injury,the more increasing of the expression of tTG. Changes in renal tTg occur in progressive chronic renal diseases in humans independently of the original etiology .tTg may therefore play a role in the pathogenesis of renal fibrosis in patients with CKD and can therefore be considered a potential therapeutic target. In Lupus nephropathy,IgAN and Nephritis of anaphylactoid puerperal, tTG staining correlated with urinary protein excretion. The urinary protein excretion can be used to estimate the development of chronic Kidney disease. In the LN group, C3,C4 can be used to estimate the development of Lupus nephropathy.
