Objectives: Apply respiratory tract quiet to use narcotic, establish animal model, detect content of ALT,AST,LDH, and observe the morphological injury of Liver and kidney by the Petroleum Bitumen fume in mouse.Methods: 200 Kunming mice were provided by Laboratory Animal Center of University. They were 4 weeks old and half were male. All the mice were randomly divided into experimental and control groups. Each group had 40 mice. The experimental groups were exposed to bitumen fumes (average 41mg/m~3, 82 mg/m~3 and 165 mg/m~3) by inhalation in a sealed vessel for 2 hours/day, 6days every week. One third of the mice were executed after inhale bitumen fumes for 2W, 4W and 8W. The weight of the mice was measured once a week. Morphologic features of liver and kidney tissue were observed by optical microscope and electron microscope. The content of AST, ALT and LDH were measured by complete automatism analyze instrument.Results: (1) Effect on growth: Bitumen fume has negative effect on the growth of the animals. The body weight gains of the animals in bitumen fume-treated were lower than in the control groups, significantly on the high treated group. (2) Effect on content of ALT,AST,LDH: With the exposure level (mean 41mg/m~3,82 mg/m~3,165 mg/m~3) increasing and the time (2W,4W,8W) lasting, content of ALT,AST,LDH is not significant. (3) Effect on liver and kidney tissue morphology: With the exposure level (mean 41mg/m~3,82 mg/m~3,165 mg/m~3) increasing and the time (2W,4W,8W) lasting, the Bitumen fume could result in the injury, which included the mitochondria swollen, chromatin margination, pyknosis etal in liver and kidney cells in the high group of 2W and any kind of groups of 4W and 8w.Conclusion: Bitumen fume can cause negative effect on the mice growth and development. A long-time exposure of bitumen fume can cause the injury of liver and kidney tissue .
