Objective To investigate the changes of transcription factor nuclear-κB (NF-κB), TNF-α, IL-18, MPO in lung and Serum Ca2+, in order to explore the effects of reduced glutathione(GSH).Methods 72 Wistar rats were divided into 3 groups by the random datatable : ANP group, GSH pretreated group, and SO (sham operation) group. Rats of ANP was induced by retrograde injection of 5% sodium taurocholate into the pancreatic duct. GSH pretreatment group were administrated into muscle at the dose of 0.15g/kg. Animals in three groups were killed at 6, 12and 18h after induction of the model and blood, lung tissue, pancreas tissue were measured. The activity of IL-18, TNF-αin lung tissue were measured by immunohistochemistry. Serum Ca2+ and MPO in lung tissue were determined by chromatometry. Intrapulmonary expression of NF-κB p65 was detected by RT-PCR. The histopathology of lung was observed under the light microscope.Results Intrapulmonary expression of NF-κB mRNA is higher in ANP group than GSH pretreatment group, and the latter higher than SO group at 6h. the activity of IL-18, TNF-α, the level of MPO in lung tissue and serum Calcium were much higher at 6h, 12h, 18h in ANP group and GSH pretreated group after the induction of pancreatitis, with statistical significance compared with the SO group (p< 0.05). After treatment with GSH, the level of IL-18, TNF- a become lower at 6h, 12h, 18h and MPO at 12h, at the same time serum Calcium became higher.Conclusion 1. The activity of NF-κB and the level of TNF-α, IL-18, MPO havesomething to do with acute lung injury in experimental acute necrosis pancreatitis.2. GSH may inhibit the activity of NF-κB and neutrocyte in lung tissue, decrease the concentrations of inflammatory cytokines (TNF-α, IL-18). SoGSH should be useful for the treatment of lung injury.
