Cerebrovascular disease (CVD) is a devastating clinical condition,which severely do harm to human healthy.The incidence and mortality from CVD have the tendency of increasing with life span elongation.CVD is the third leading cause of death in our country and even more is the first leading cause in some areas. 75% survival patients were severely disabled which bring heavy distress and economic loss for himself,the family and the social.Ischemic cerebrovascular disease (ICD) is the central nervous system disease caused by the artery block temporary or permanent.It approximately hold 80% in the cerebrovascular disorders. Currently thrombolysis is the main therapy method ,but there are some dangers and restrictions for hemorrhage and reperfussion injury. Cerebral ischemia-reperfusion (CIR) injury is the damage even severely when the blood stream recanalize in the ischemic brain tissue which has experenced cerebral ischemia for some time.Therefore investagate the mechanism of the cerebral ischemia injury and actively develop the protective drug for the damage have very important theoretical significance and utility value.Currently it is widely confirmed that nerve cell experienced two death style:necrosis and apoptosis after focal cerebral ischemia. Necrosis usually take up in the region that ischemia occurrences very fast and very severely, but apoptosis occurrences in the region oflight ischemia. The damages of cerebral ischemia are cerebral infarction and reperfusion that blood stream recanalize. The region that affected by reperfusion injury is ischemia penumbra region. The main pathologic change is apoptosis in the area.Our research team has done a lot of studies on NaoxinKang Injection.The data of our research team has showed that NaoxinKang Injection has the effect of improving arterial blood streaming and ameliorating the blood viscosity, anti-thrombotic effect, inhibiting platelet aggregation. But we don,t know wether NaoxinKang Injection has protective effect on cerebral ischemia and how.Its mechanism need continue to be investigated.The aim of this work is to observe the protective effects of NaoxinKang Injection on cerebral ischemia-reperfussion injury and its possible mechanism on the level of in vivo through setting up incomplete cerebral ischemia and cerebral ischemia- reperfusion model in rats.Thus can provide a theoretical basis for better, safer use and targeting application of this therapy in clinical treatment. Through setting up incomplete cerebral ischemia and cerebral ischemia- reperfusion model in rats, the effects of NaoxinKang Injection on the level of SOD, MDA, NO,CK,LDH both in the brain tissue and serum of rats.,and the neurologic deficit score were detected. At the same time, samples were taken to store in the fixation liquid for HE.The results of the experiment showed that NaoxinKangInjection could significantly reduce the content of CK and LDH,wich indicates that NaoxinKang Injection could relieve the damageand protect the integrity of cellular membrane. NaoxinKang Injection play a role of cellular membrane stability .Meanwhile NaoxinKang Injection could obviously decrease the content of MDA and could increase the activity of SOD both in brain tissue and serum; and reduce the infarction area. It also could significantly increase the neurologic deficit score .This indicated that NaoxinKang Injection could increase the anti-oxidation activity of the enzyme, enhance the ability of removing free radicals and reduce the free radical from ischemia-reperfusion, so that it improve function of brain. The results of the experiment also demonstrated that NaoxinKang Injection could strickingly reduce the content of NO in brain tissue ,which showed that NaoxinKang Injection could ease the harm of NO.Based on the above experiments , it has been proved that NaoxinKang Injection increased the ability of removing free radicals, decreased lipid peroxidatic,protect both integrity and stability of cellular membrane,inhibited production of NO, and then protected the neurons from hypoxia damage.
