| ObjectivesThe sanitation and safety of drinking water will influence the health of user. Cholorination is the present most method of sterilization in each nation. The cholorination of drinking water is the great project, which is related to each of our daily life and health, because of its broaden influence, it can attract each of our attention. Since the Disinfection By-products have discovered the harmful function of health, most studies in domestic and overseas had done more researches on that, including the form mechanism of co-product, the conditions of influence, and the studies in the influences of body health, et al. The present study makes source water and strainer periph water coming from the same water plant respectively at the period of dry season and abundant season as the subject, using the test animals to observe the trauma of hepatic cells, didymus cells and HL-7702 cells from the organism attractive of source water and tap water. With the comparison with source water and tap water, it will be taken as the guide of water treatment and provided the foundation for evaluating the water quality.MethodsTo collect the source water and strainer periphery water respectively 200L at the same tap water factory in the abundant and dry water periods, after water sample precipitated, organic extractions of surface water and tap water is enriched by XAD-16 macroporous resin. The test animals were randomized into four groups, the dosage of narcotics is: negative group (0.25 ml·kg-1DMSO-A group), low-dose group (1.25 L·kg-1-B group), middle-dose group (2.5 L·kg-1-C group), organic extractions high-dose group (5 L organic extractions of drinking water?kg-1-D group). Each group has ten animals (male 5 and female 5), treatment groups were given by mouth, respectively for 15 days, then weigh the animals, and do the experiment about liver lipid peroxidation and didymus toxicity test.Making liver cell and didymus cell suspension, respectively putting 10ul DMSO(A group), 10ul DBPs extract, making the final concentration as each 1 ml culture fluid contain 35 ml(B group), 70 ml(C group), 140 ml(D group) of untreated water, mixed it up, put it to the 37℃thermostatic waterbath 2 hours, finish the single cell gel electrophoresis.Using the HL-7702 cell to lay the narcotics density according to cell survival rate exceed 90%. Making DMSO as the solvent control(A group), respectively put the 10 ul DBPs extract, making the final density as each 1 ml culture fluid having 7 ml(B group), 14 ml(C group), 35 ml(D group) of untreated water. Using TBA to detect the MDA, and using DTNB direct method to detect glutathione perhydride.Results1. Organ indicesD group of liver index of female mice by dyed the poison of the organic extractions of tap water at plentiful water period is lower than C group obviously, C group of liver index of female mice by dyed the poison of the organic extractions of tap water at withered water period is higher than A and B group obviously, there are no obvious difference in other groups.There are no obvious differences in groups of liver index of male mice by dyed the poison of the organic extractions.C and D group of testis index of male mice by dyed the poison of the organic extractions of surface water at withered water period is higher than B group obviously,there are no obvious difference in other groups. 2. Enzyme activity in testis cellThe LDH activity of organism extractive of experimental groups from source water coming from abundant and dry seasons is significant lower than that in control, and The LDH activity decreases with the narcotics dosage increases. The LDH activity of C and D group are significant lower than that in control group. The LDH activity decreases as the narcotics dosage increases.With the narcotics dosage of source and tap water coming from abundant season increases, the G-6-PD activity also increases. With the narcotics dosage of source and tap water coming from dry season increases, the G-6-PD activity decreases.3. SCGE(1) Experiment in vivoEach narcotics group has damage on DNA of testis cell, and as the dosage increased, the propotion of DNA damage also increases. B and C group of tap water coming from abundant season emerge III and above III DNA damage, but the same group of tap water coming from dry season did not emerge that phenomenon.(2) Experiment in vitroEach narcotics group has greatly damage on testis cell, there is no significant difference among any dosage group of tap water. As the dosage of tap water coming from abundant and dry season increased, the propotion of DNA damage also increases. There is no damage on DNA of liver cell in organism extractive groups.4. lipid peroxidationEffects of lipid peroxidation of female mice by the organic extractions in abundant season: MDA of D group in tap water is lower than C group, GSH-PX of B group is higher than A,C,D group; MDA of B,C,D group in source water are lower than A group, The SOD of source and tap water narcotics group are obvious lower than A group, also with the dosage increased, the SOD activity decreases.Effects of lipid peroxidation of female mice by the organic extractions in dry season: MDA and GSH-PX of C and D group in tap water are higher than A and B group, SOD of C and D group are lower than A and B group. MDA,GSH-PX and SOD of D group in source water are lower than A,B and C group.Effects of lipid peroxidation of male mice by the organic extractions in abundant season: MDA of C group in tap water is higher than A,B and D group,GSH-PX of B group is higher than A group ,GSH-PX of D group is lower than B and C group, SOD of D group is lower than SOD of A,B,C group,MDA , GSH-PX and SOD of narcotics group in source water are higher than A group, with the dosage increased, MDA , GSH-PX and SOD increase first then decrease.Effects of lipid peroxidation of male mice by the organic extractions in dry season: MDA of A group in tap water is higher than narcotics group, GSH-PX of narcotics group is lower than A group, also with the dosage increased, GSH-PX decreases. SOD of narcotics group are higher than A group, also with the dosage increased, The activity of SOD decreases. MDA of B group in source water is higher than A,C and D group,GSH-PX of narcotics group is lower than A group, also with the dosage increased, GSH-PX decreases. SOD of A group is higher than C and D group, lower than B group, also with the dosage increased, The activity of SOD decreases.Effects of lipid peroxidation of HL-7702 by the organic extractions in abundant season: MDA of B group in tap water is lower than MDA of A and C group, MDA of D group is lower than MDA of A,B and C group; GSH-PX of B is lower than GSH-PX of A, C, D group, MDA and GSH-PX of B, C, D group in source water are higher than that of A group. Effects of lipid peroxidation of HL-7702 by the organic extractions in dry season: MDA and GSH-PX of narcotics group in tap water and MDA of narcotics group in source water are all decrease with the dosage increased, GSH-PX of narcotics group in source water are higher than A group, with the dosage increased, GSH-PX increase.Conclusion1. The organism extractive of drinking water at the dry water period and the abundant water period contains material of lipid peroxidation function to rat liver system and HL-7702 cell, and after the metabolism of the organism extractive of drinking water, and liver still contains material of lipid peroxidation, and it probably contains material of direct injury to cell in the organism extractive of tap water after cholorination.2. The organism extractive of drinking water at the dry water period and the abundant water period have vir-repression to reproduction system, The enzyme activity of LDH and G-6-PD changes a lot in didymus of male rat.3.When the dosage of organism extractive increase, the trauma of didymus cell of rat was been aggravated, this phenomenon will be manifested differently in source water by comparison with tap water.4.At the same condition of extraction, the dosage of organism extractive of drinking water at the dry water period is more than the drinking water at the abundant water period, also has more obviously vir-repression to reproduction and endocrine system. |
