Objective: To evaluate therapeutic effects and side effects of GnRH-αreceptor agonist Leuprorelin and gestrinone on endometriosis.Methods: Randomly choose 80 cases of patients with recurrent endometriosis from June of 2004 to June of 2006, diagnosed through abdominal speculum and pathological analysis in the past 5 to 10 years. Divide the patients into two groups according to clinical stage (CS) standard. Each group were identically given GnRH-αreceptor agonist Leuprorelin and gonadal hormone resistance medicine gestrinone, before comparing differences in symptom releasing extent, uterus volume changes through abdominal ultrasonic detection, E2,FSH,LH value and ECG, routine blood/urine changes, as well as the menstruation variations before and after the trial. All side effects were under surveillance. A retrospective analysis was performed through related clinical data.Results: The algomenorrhea release rates for Leuprorelin group and gestrinone group were 91.46% and 64.98%, respectively.While the amenorrhea rates were 92.5% and 62.5%, respectively. Slight changes could be viewed in E2, FSH and LH values between the two groups, without obvious climacteric symptoms. Volumes of endomembrane ectopic focus neoplasm decreased by 78.03% and 61.33%, respectively, with a significant difference between two groups (p<0.01). A higher side effect occurrence was observed in gestrinone group. No remarkable changes in ECG, routine blood/urine detection, liver and renal function. In gestrinone group, 6 cases of acne were observed and 4 other cases with body weight increased. However, these patients recovered from such disorders 2-5 months after the drug withdrawal.Conclusion :1. Ultrasonic detection is more practical in prognosis of uterus endometriosis.2. Both Leuprorelin and gestrinone can effectively relieve symptoms such as algomenorrhea, to cause amenorrhea. However, Leuprorelin group demonstrated better effect than gestrinone group.3. Both drugs can decrease volumes of endomembrane ectopic focus neoplasm. And Leuprorelin group demonstrated better effects.4. No significant changes could be viewed in E2, FSH and LH values for both groups after drug administered, with rare side effects on autonomic nerves.5. Without any reverse-adding treatment, all patients showed better tolerance to a continual 3 months therapy with GnRH-αreceptor agonists.6. Gestrinone can induce more side effects than Leuprorelin.
