| Tumor, especially malignant tumor has been one of the most serious diseases for many years. All over the world, about 7 million patiens have died from malignant tumors every year. The experts of the world try their bests to study and develop the medicines of treating malignancies. Above all, the traditional Chinese medicine is preference because of its well therapenutic efficacy and minor-toxicity. Mistletoe, semi-parasitic plant, is an endemic traditional Chinese medicine of Ji Lin province. In the past, it was used to eliminate beriberoid disease, reinforce liver and kidney, strong bones and muscles, anti-abortion, drop blood pressure, resist virus, subdue blood sugar, and so on. Recent years, the anticancer effects of mistletoe extracts attracted generally attentions of scholars. Its finished products had been present in Europe, the commodity was named Iscardo, used to treat tumors. In Korea, Progresses about anti-tumor effects of the Lectin from mistletoe were got, but our nation had no systematic investigation reports about the anti-tumor actions of mistletoe alkali.Adenoid cystic carcinoma is a malignant tumor, mainly located in the salivary gland. Surgery and radiation are traditional therapies. Because its high-grade malignancy and post-treatment recurrence, many toxic and adverse effects after western medicine chemotherapy, up to days, there was no completed curative effects. In this experiment, we mainly approached theinhibitory action and mechanism of the mistletoe alkali on the adenoid cystic carcinoma(ACC) cells (from salivary gland).In virto, CCK-8 assay and growth curve were utilized to investigate the inhibitory actions of alkali on the growth of ACC cells. Next, we studied the effects of alkali on the cell cycle and the activity of proliferation of the ACC cell through the PCNA immunohistochemical staining. We also used the AO/EB fluorescent staining and Caspase3, Caspase8, Caspase9 immunohistochemical staining to observe the apoptotic influences and mechanism of mistletoe alkali on ACC cells. In addition, used the invasive model-boyden chamber assay to study the influence of alkali on invasive capability of ACC cells.Experiment results demonstrated: (1)In vitro, mistletoe alkali inhibitated the growth of ACC cells and the IC50 value was 2.24μg/ml; (2) PCNA immunohistochemical staining displayed that the expression of PCNA of alkali groups was inferior than the control groups; (3)AO/EB fluorescent staining demonstrated that alkali could induce the apoptosis of ACC cells; (4)Caspase3, Caspase8, Caspase9 immunohistochemical staining showed that the expression of alkali groups was higher than the control groups. so, we could say, mistletoe alkali could induce the apoptosis of ACC cells through promoting the apoptotic factors; (5)In invasion assay, the number of ACC cells of alkai groups invaded the recombinated basement membrane obviously fewer than of negative groups.We could make a conclusion from the results: the mistletoe alkali was able to inhibit the growth of ACC cells, mainly through inhibiting the proliferation and inducing the apoptosis of them. meanwhile, mistletoe alkalicould decrease the invasive capability of ACC cells. The research, development and utilization of mistletoe alkali will establish a new perspective for the clinical treatments on malignant tumors. |
