ObjectiveGlioblastoma multiforme is the most common and lethal primary malignant brain tumor. Although considerable progress has been made in surgical technique and radiotherapy for brain tumor patients, the impact of these advances on clinical outcome has been disappointing, with median survival time not exceeding 15 months. It is significant to investigate the biological features of this disease.Cysteinyl leukotrienes (CysLTs, including LTC4, LTD4 and LTE4) are 5-lipoxygenase (5-LOX) metabolites of arachidonic acid. The elevation of CysLTs and the expression of CysLT receptors, including two subtypes CysLT1 and CysLT2 ,have been demonstrated in glioma cell. However, the relationship of CysLTs receptor and glioblastoma is not yet know. The dependablity between the expressions of CysLTs receptor and malignant progression of glioblastoma is unknown. The studies on the expression of CysLT1 and CysLT2 is promising to bring a new breakthrough in the diagnosis and therapy of gliomas. Thisexperiment determined the expression of CysLT1 and CysLT2 on 48 patients of glioblastoma and 3 normal brain tissue. The objective was to examine the relation between the expression of CysLT receptors and malignant progression of glioblastoma and to find new method to heal glioma.Material and MethodsThe glioma samples were collected from 48 patients (33 male, 15 femal, mean age 44.5Y, range from 5 to 78Y) during the period between 2002.8 and 2003.8 in the Second Affiliated Hospital, School of Medicine, Zhejiang University. According to WHO2000 CNS tumor stage, the samples were divided into low-grade group(grade I~II) and high-grade group(grade III~TV). Low-grade group included 31samples(21 male, 10 female, mean age 41.6Y, range 5-64), high-grade group included 17 samples(11 male, 6 female, mean age 49.8Y, range 29-78). 3 samples of normal brain tissue were collected as a control group, which were got from the patients with deep-part benign tumor during approaching the tumor.Tissues were performed to detect the expression of CysLT1 and CysLT2 with immunohistological technology. The index of immunological activity of these samples was accounted by semiquantitative scoring methods, and statistical analysised with SPSS11.0.ResultsThe cell with CysLT1 positive expression represents the yellow cytoplasm, butalso can represent on the nuclear membrane in several high-grade glioma. CysLT1 immunostaining was intense in the microvascular endothelial cells but weak in other cells in normal brain tissue. CysLT1 was robustly induced in astrocytoma and gliocytoma cells, increased in the microvascular endo-thelial cells, and also express in the neuron-and glial-appearing cells around the tumor. The index of immunological activity is depend on the malignant progression.CysLT2 immunostaining was intense in the smooth muscle cell of the arteriole and venular, but absent in endothelia cells and also absent in the microvascular endothelial cells. CysLT2 was absent in the centre of the tumor, but robustly induced in the neuron-and glial-appearing cells around the tumor. The index of immunological activity is depend on the malignant progress.ConclusionsCysLT2 and CysLT1 were robustly induced in the glioma, and the index ofimmunological activity is depend on the malignant progress.CysLT2 and CysLT1 were different in the expression location and intension in theglioma.
