Twenty recurrent aphthous ulcer(RAU)patients were choose for this experiment .We collected different samples from them including saliva, exfoliated cell, biopsy samples and blood for real-time fluorescent quantitative PCR(FQ-PCR) test. In this way we can compare the quantities of virus in different samples and give more explanation of virus causes of RAU.Background and Objective: CD4/CD8 is lower in RAU patients. All its recurrent characteristic symptoms that looks like herpes infection and intendance to infect epithelium only imply that the cause of RAU maybe viruses. The objective of researches are all human herpes virus (HHV) recently.There are 9 kinds of HHVs. They are separated into three groups. At present ,the researches about RAU involve HHV1-8 and three groups.αherpes virus (for example HSV, VZV) can reproduce rapidly and induce to cell disease. They can live in many kinds of hosts. They can infect sensory nerves also.βherpes virus (for example HCMV) has not many hosts. The period of virus is longer. They can stay in salivary gland, kidney and so on and lead to the latent infection.γherpes virus (for example EBV) have fewer hosts. Most of the aim-cells are B cells. They can stay in them for very long time.The results of researches about RAU involve HHV1-8. For instance, it is reported that many viruses have been found in RAU patients as following HSV-1, VZV, EBV, HCMV, HHV-6, HHV-7, HHV-8, and the difference isnotable in statistics, but some researches also find opposite results. Comparing the different experiments, the different ways of operations and collection of samples maybe the keypoint. The aim of this study is to choose real-time fluorescent quantitative PCR to test HCMV which has many opposite results and HHV-8 which has fewer results now .Methods: We collect RAU patients in the second hospital, plastic surgery and dental clinic of Dalian medical university. 18 RAU patients, 6 male with an average age:44 years, 12 female with an average age: 26.7 years. Samples like saliva, exfoliated cells, biopsy samples and blood were collected, at the same time we choose 10 health controls,saliva, exfoliated cells and blood were collected from them and we took biopsy samples from 10 patients who had done extraction therapy, we took saliva and exfoliated cell from 3 patients who is going to do extraction therapy then we took biopsy samples from them when they finished their therapy. Preserve and take the DNA from them in biochemistry department of Dalian Medical University and samples were sent to TaKaRa company for test.Results: The result of HCMV test: Destination viruses were detected in 8 out of 70 (11.4%) samples from 5 out of 16(31.25%) patients with RAU and biopsy samples from those 5 patients are all positive, 5 samples(3 blood samples and 2 saliva samples) from 3 healthy controls are positive. 2 blood samples show positive in test account for 11.1% of all the blood samples. 1 saliva sample is positive ,account for 5.6% of all the saliva samples. The result of HHV-8 test: apply real-time fluorescent quantitative PCR to test the patient samples and the control samples at the same time. That show no result, so we can say that the samples are negative or there are very few viruses in them.Conclusions: Exact probability test showed that the HCMV positive rate in samples and patients of RAU group has no significant statistics difference with their health controls. The HCMV positive rate in saliva, exfoliated cell, biopsy samples and blood samples has no significant statistics difference with their health controls also, but the HCMV positive rate in biopsy samples has significant statistics difference with their health controls. The result imply that the viruses maybe latent in blood and saliva gland, when the immunology of patients go down the viruses can generate inoral mucosa and cause RAU.
