Autologous fat grafts have many advantages in the reconstruction for soft-tissue and contour defects. Fat is soft , pliable, readily available. However , the successful use of fat grafting is frequently limited by the sometimes low survival rates. Many experts did some research works to improve the survival rate. Ang-1 and bFGF have definite promoting effect when they were used alone. But until now ,there is no report about effect of combined treatment with Ang-1 and bFGF in fat particle auto-transplantation.Objective: Through establishing a rat fat-transplantation model, injecting Ang-1 and bFGF into fat particles, with the function of Ang-1 or bFGF alone to improve angiogenesis and fat graft survival rate , to investigate the effect of combined treatment with Ang-1 and bFGF in fat particle auto-transplantation.Method: Twenty-four female Wistar rats weighting 290-310 g were randomly assigned into four groups and treated as follows:Grafts in 1st cephalic side were mixed with normal saline as control groupsGrafts in 1st cauda side were mixed with 2μg bFGFGrafts in 2nd cephalic side were mixed with 0.25μg Ang-1Grafts in 2nd cauda side were mixed with 0.25μg Ang-1+2μg bFGFGrafts in 3rd cephalic side were mixed with normal saline as control groupsGrafts in 3rd cauda side were mixed with 2μg bFGFGrafts in 4th cephalic side were mixed with 0.25μg Ang-1Grafts in 4th cauda side were mixed with 0.25μg Ang-1+2μg bFGF1 ml×2autologous fat grafts were excised from abdominal cavity of every Wistar rats.Then it was purified and mixed with 2μg bFGF,0.25μg Ang-1,0.25μg Ang-1+2μg bFGF and NS respectively .Finally transplanted them in the rats'dorsums. The 1,2 group transplanted tissue were taken out after 7 days and were stained with CD34 immunohistochemistry method. The microvessel density was measured by medical image analysis system. The 3,4 group transplanted tissues were taken out after 30 days, and their volumes were measured with their suvival rate being calculated.Results: The 1,3 groups'microvessels were brown color after CD34 staining. The values of microvessel density in all treated groups were significantly higher as compared with in saline group. The values of microvessel density in Ang-1 and bFGF group was significantly higher than in Ang-1 group or in bFGF group. There was no significant difference between Ang-1 group and bFGF group. The 3,4 groups'survival rate was calculated.The survival rate of all treated groups was significantly higher as compared with saline group. The survival rate of Ang-1 and bFGF group was significantly higher than Ang-1 group or bFGF group. There was no significant difference between Ang-1 group and bFGF group.Conclusion: Combined treatment with Ang-1 and bFGF in fat particle auto-transplantation is a more effective method than with Ang-1 or bFGF alone.
